Isolation,Purification,Structural Elucidatuion,and The Use In Cigarette Of Polysaccharides From Polygonatum Odoratum

There are four parts in this research including determination of optimum extraction conditions of ultrasound-assisted technology of Polygonatum Odoratum polysaccharide,purification,physicochemical property research,structure elucidation and the moisturizing property in cigarette.With the univariate experiments and response surface methodology,the optimum ultrasound-assisted extraction techniques were confirmed:the ratio of water to material 50,ultrasonic power 426 w,extraction time 35 min,and the POP extracting yield reached 29.29%under the conditions.The crude polysaccharide of P.Odoratum after ethanol precipitation was isolated and purified using DEAE Sapharose Fast Flow and Sephacryl S gel-filtration chromatography repeatedly.Four water-soluble polysaccharides(POP1-POP4)were obtained,three of them were determined to be homogeneous polysaccharides(POP2-POP4)by HPLC and UV.IR spectra showed that the sugar characteristic groups were existed.Correlation with the retention time of HPLC and calibration curve of dextran standards,their molecular weight were less than 1.0×103Da,more than 2×106 Da,2.5×103 Da from POP2-POP4,respectively.With the analysis of physical-chemical character and practical experience,POP4 was most likely to be the active component.So,the structural analysis of POP4 were carried by spectral technologies include-ing IR,GC,GC-MS,and NMR to identify the sugar composition,the number of sugar residues and the glycosyl residue linkage.POP4 was a neutral polysaccharide and composed of fructose mainly,Methylation analysis showed it contained 2,6-and terminal fructose linkage.NMR analysis further indicated that the sugar residue were existed as βconfiguration and the configuration was furanose.In conclusion,POP4 was a soluble(2,6)-linked β-D-Fructan with the furanose configuration.The study on the application of cigarette showed:P.Odoratum polysaccharide plays a good moisturizing effect.It can reduce the loss of tobacco moisture,improve the comfort of smoking,and it makes less influence on security of the cigarette.
