Our country is diosgenin producing countries,most companies sti ll using the direct acid hydrolysis of diosgenin preparation,and its huge amount of acid produced large amounts of sewage has been criticized.The compound enzyme hydrolysis method,with mild conditions,less sewage discharge,diosgenin high yield charact eristics,is a clean production process.In this paper,the purpose of comprehensive development and utilization of Dioscorea zingiberensis was to decompose starch and cellulose and other impurities in Dioscorea zingiberensis by compound enzyme hydrolysis,and the extraction of diosgenin was carried out.The wastewater produced in the extraction process was treated,The waste liquid in a large number of carbohydrates into 5-hydroxymethyl furfural;Finally,diosgenin in vitro anti-free radical activity research,the main research content is as follows:?1?Research diosgenin direct acid hydrolysis extraction.The optimum acid hydrolysis conditions were determined as follows: 2 mol/ L sulfuric acid solution,acid hydrolysis at 90? for 4h,and the yield of diosgenin was 1.76% under the condition of single factor experiment.The extraction process of diosgenin from the direct acid solution was simple,but the acidity was larger,and the side reaction was more in the violent reaction condition,and the yield of diosgenin was lower.?2?Study on Extraction of Diosgenin by Compound Enzyme Hydrolysis.The optimum enzymatic hydrolysis conditions were determined by single factor experiment.The optimum enzymolysis conditions were as follows: 0.5% compound enzyme was adjusted,p H was 5.5,hydrolyzed at 50? for 8h,acid hydrolysis was carried out after enzymatic hydrolysis,and the optimum conditions were determined by s ingle factor experiment: 1 mol/L concentrated sulfuric acid,at 90? for 4h;Finally,single factor experiment and orthogonal experiment to determine the optimal hydrolysis of diosgenin by compound enzyme hydrolysis method: adding 0.5% compound enzyme,at p H = 5.5,50 ? under the conditions of enzymatic hydrolysis 8h;adding concentrated sulfuric acid and distilled water,the sulfur ic acid concentration of 1mol/L,at 90? for 4h;petroleum ether extraction,in the feed ratio of 1:8,60? extraction 3 times The Under these conditions,the yield of diosgenin was 2.56% and the purity was 92.85% by HPLC.?3?Study on the Anti-free Radical Activity of Diosgenin in.First,the electrochemical behavior of diosgenin was studied by cyclic voltammetry using multi-walled carbon nanotubes modified electrode.Secondly,the effect of diosgenin on DPPH free radical,hydroxyl radical and superoxide radical was studied by cyclic voltammetry.The effects of diosgenin on DPPH radical,hydroxyl radical and superoxide radical were calculated by data.clearance rate.The results showed that the half inhibitory concentration?IC50?of diosgenin on DPPH free radicals was 0.667mmol/L,the half inhibitory concentration?IC50?of diosgenin to hydroxyl radical was 0.714mmol/L,diosgenin The half inhibitory concentration?IC 50?of the oxygen free radical was 1.048 mmol/L.The results showed that diosgenin had certain antioxidant activity,and the removal efficiency of DPPH free radical was the best,and the hydroxyl radical was the second,and the scavenging effect of superoxide radical was poor.?4?Preparation of 5-Hydroxymethyl Furfural?HMF?by Fructose Syrup Catalyzed by Cr Cl3/PVP Nanofiber Catalyst.Cr Cl3/PVP nanofibers were prepared by electrospinning,and characterized by thermogravimetric differential thermal analysis,infrared spectroscopy and scanning electron microscopy.Firstly,HMF was prepared from glucose as raw material.In the reaction system of tetr aethylammonium chloride,the calcination temperature,catalyst addition amount,catalytic reaction temperature and catalytic reaction time were selected to prepare 5-hydroxymethylfurfural Yield effect.The experimental results show that the Cr Cl 3/ PVP nanofibers calcined at 200? have a good catalytic effect.When the reaction temperature is 120?,the reaction time is 20 min and the amount of catalyst is 10%,the catalytic activity of HMF is catalyzed by TEAC as catalyst.The yield was 65.6%.Finally,5-hydroxymethylfurfural was prepared by catalyzing fructose syrup using Cr Cl3 / PVP nanofiber catalyst.The optimum reaction conditions were determined by single factor experiment and orthogonal experiment using Cr Cl3/PVP nanofiber membrane as catalyst and TEAC as solvent.The reaction temperature was 100?,the reaction time was 30 min and the amount of catalyst was 10%.The average yield was 38.94%.
