Fluorescent probes are a kind of excellent detection tools for fluorescent imaging,Due to the properties of high sensitivity,good selectivity,low cost as well as simple operation,which lead to the widely use of fluorescent probes in biochemistry,materials,analytical and environmental chemistry and other fields.Palladium,one of the noble metal,was widely used for fuel cell,jewelry,dental materials,pharmaceutical and electrical equipments.Also,Palladium is ideally suited for catalysts(chemical synthesis catalysts and automobile exhaust catalysts).However,the extensive use of palladium increases the risks associated with health hazards.The proposed Maximum dietary intake of palladium is no more than 15 μg per person per day.Consequently,effective methods to detect trace palladium are necessary.Cysteine is an important a-amino in human body,which plays important roles in physiological and pathophysiological process.Therefore,it is of great significance to develop highly selective and sensitive fluorescent probes for detecting the distribution and fluctuation level of Cys in vivo,which is associated with disease prevention and clinical medical research.This work mainly covers the following two parts:1.A new probe is synthesized for the detection of palladium ions.The fluorophore of the probe is the derivative of 4-Chloro-7-nitrobenzofurazan and the acrylate group as the sensitive recognition site for probe.The probe was characterized by 1H NMR、13C NMR、EI-MS and HRMS.With the 485 nm UV light irradiation excitation,the probe showed faint fluorescence.After the probe reacted with Pd2+,it glows bright green fluorescence and the maximum emission wavelength was 555 nm.The probe has a high stability,high selectivity at physiological pH conditions.The fluorescence intensity is dependent on the concentration of Pd2+ with the linear response range being 1-2 μmol/L,with the detection limit being 0.17 μmol/L.More important,the probe is suitable for fluorescence bioimaging of palladium ion in living cells.2.A new probe is synthesized for the detection of cysteine.The fluorophore of the probe is the derivative of acetonaphthone and acrylate group as the sensitive recognition site for probe.The probe was characterized by 1H NMR、13C NMR、El-MSand HRMS.With the 375 nm UV light irradiation excitation,it glows bright blue fluorescence and the maximum emission wavelength was 455 nm and the fluorescence intensity of the probe tends to stable in 28 minutes.The probe has a high stability,high selectivity at physiological pH conditions.The fluorescence intensity is dependent on the concentration of Cys with the linear response range being 1-10 μmol/L,with the detection limit being 0.10 μmol/L.In addition,the probe is suitable for fluorescence bioimaging of cysteine in living cells.3.From summarizing the research results of this thesis,it is indicated that the palladium ions and Cys probe has a high stability,high selectivity at physiological pH conditions.The synthesis of palladium ions and Cys probe provides certain ideas and theoretical reasons for the future design and research.
