Juglans regia is the direct ancestors of cultivated walnut, it belongs to the walnut, walnut genera plants. In our country, mainly live in xinjiang yili gongliu of wild walnut groove, it is extremely rare and precious wild natural resources. But it is about in the genetic linkage map construction and use of molecular marker assisted breeding, obviously lags behind other wild fruit tree species, at present, the domestic has not been reported about the map construction. Using DNA markers to construct Juglans regia molecular linkage map, will conduct relevant agronomic traits for Juglans regia in fine mapping, QTL genetic and MAS selective breeding and other scientific basis.This study use Juglans regia in the wild walnut ditch female parent and male parent and the F1 hybrid construction group, using SSR and SRAP applicable to Juglans regia reaction system and the analysis of the separation of the tags etc was studied, constructing the first Juglans regia molecular linkage map in xinjiang.And the plant height, stem diameter, maximum leaf length and minimum leaf width, etc properties for the study of correlation analysis. The concrete research content is as follows:1. This study selected two tree growing better and phenotypic traits differences of wild trees(male parent:flat round walnut, female parent:ellipse walnut) for artificial pollination, success in the 368 F1 generation of individuals. Determined at seedling stage and the F1 generation of relevant quantitative traits, and individuals for further construction.2. Screening SSR and SRAP primers, using parents and F1 of 115 SSR markers and 100 SRAP markers for screening, selected from 115 pairs of SSR primers in a sequence of 46 pairs of polymorphic rate higher separation detection role in groups, received 70 polymorphic loci. From 100 pairs of SRAP primer combinations selected 17 for augmentation with clear good SRAP primer combinations, a total of 39 polymorphism markers loci.Test a total of 109 polymorphic loci detected for subsequent composition analysis.3. Using joinmap4.0 analysis software for linkage map construction of 109 marker loci, containing 29 SSR, 33 SRAP markers of a tag in Juglans regia genetic linkage map, the map involved 15 linkage group, the total length of the genome is 757.5c M, the length of each chain group of 22.1c M-151.7c M, the average figure between distance of 12.22 c M, the average figure between each ranged from 2-13 markers on the linkage group,partial separation of tag 13, accounting for 21%.4. The study randomly selected 368 Juglans regia resources, and its phenotypic traits, SSR and SRAP genetic diversity, genetic relationship analysis testing and evaluation. 8 values of the phenotypic traits spread in the group and have a better fitting with normal distribution curve. The variable coefficient shows that the degree of variation of each character is more than 10% and has a larger variation. Rich variety of genetic variability provides theories for genetic improvement and its molecular breeding of Juglans regia.
