Cucumber seed oil has the effect of beauty and health care.Development and utilization of cucumber seed oil,not only for natural cosmetics research and development to open up a new path,it can also be used as a green food additive,sodevelopment prospects.Currently,people pay more and more attention to beauty and health care,cucumber seed oil application prospects are more broad.In this study,35 different ecological type of cucumber germplasm resources were used as test materials,soxhlet extraction was used for seeds crude fat,evaluated and analyzed seed oil content of cucumber.According to the results,the variety of high oil content D1158-2(oil content was40.59%)and the variety of low oil content D0872(oil content was 32.61%)were selected as test materials,and the hybrid combination(D1158-2×D0872)was selected.The 256 F2 segregating populations formed by F1 single plant selfing were used as materials,using bulked segregation analysis(BSA)and single nucleotide polymorphism(SNP)markers for oil content related gene location,and to search for molecular markers closely linked to the oil content of cucumber seeds.The research aimed to lay the foundation for the future research on the basic application of cucumber seed oil and the breeding of its new varieties.The main results of this study are as follows:1)There were significant differences in seed oil content among different cucumber varieties,among them the seed oil content of southern China cucumber was the highest,the content was40.59%;the seed oil content of North China Cucumber was the lowest,the content was31.03%.According to the selection rate of 10%,the cucumber seed high oil content of 4 species and low oil content of 4 species were selected from the tested materials in the 35 samples.2)The order of oil content of four kinds of ecological types of cucumber was european greenhouse type > southern China type > pickled type > north China type.The correlation between the oil content of cucumber seed and the ecological type was not obvious.3)Under normal temperature conditions,the oil content of cucumber seeds remained unchanged at3 years.4)In this study,5 candidate regions related to traits were obtained by correlation analysis,among them the total length of 0.268 Mb,and total of 25 candidate genes were identified in the candidate Area,1 genes of non synonymous mutation SNP locus.5)Gene annotation in the GO database identified 4 genes related to seed oil content Csa3M912950.1,Csa3M912950.2,Csa3M912940.1 and Csa3M912380.
