[Object] Comparative study of Cyperus rotundus and third-generation aromatase inhibitor letrozole in rat ovarian P450arom,CYP17,TNF-αexpression,and to explore the role of Cyperus rotundus menstruation drug targets and its mechanism.[Method]Estrous cycle,the rules will be 48 female SD rats of clean grade were randomly divided into four groups,simultaneous sexual cycle in proestrus beginning of each group were given different drugs,each group rats were randomly divided into three batches of the principles of 5,7,79respectively in the first days of continuous medication were killed, ovaries,uterus,rat follicle count, using immunohistochemistry to detect ovarian P450arom,CYP17, TNF-αin the expression of different intervention group.[Result] In this study, the results of rat primary follicle count Cyperus rotundus group,letrozole group,Cyperus rotundus+letrozole group the average number of follicles were no statistically significant difference(P> 0.05);on rat secondary follicle count the results of Hong attached group, letrozole group,Cyperus rotundus+letrozole group the average number of follicles was higher than the control group,the differences were statistically significant(P<0.05).Cyperus rotundus respectively with letrozole group, Cyperus rotundus+letrozole group differences were statistically significant (P<0.05).In rat ovarian tissue results in the expression of P450arom visible Cyperus rotundus group,letrozole group,Cyperus rotundus+ letrozole group,the average gray value of ovarian P450arom were higher than blank control group,Differences were statistically significant(P<0.05). Cyperus rotundus respectively with letrozole group,Cyperus rotundus+ letrozole group differences were statistically significant(P<0.05).CYP17 rat ovarian tissue expression of the results of Cyperus rotundus group and blank control group,letrozole group difference was statistically significant (P<0.05),the rest were no statistically significant difference(P>0.05).In rat ovarian tissue expression of TNF-a in the results of the control group, respectively Cyperus rotundus group,letrozole group,Cyperus rotundus+ letrozole group differences were statistically significant(P<0.05),the rest did not differ Statistically significant(P>0.05).Endometrial tissue in rats the expression of P450arom was no significant difference(P>0.05).[Conclusion] 1.Cyperus rotundus,and letrozole can facilitate follicular growth in rats.2.Cyperus rotundus,and letrozole can be reduced ovarian P450arom and the expression of TNF-αin order to promote follicle development and ovulation.3.Cyperus rotundus can down-regulate CYP17 expression in ovarian tissue in order to promote follicular development and ovulation,while reducing androgen production, prevention of hyperandrogenism.4.By Cyperus rotundus with letrozole in rat ovarian tissue P450arom, CYP17, TNF-αexpression in follicle development and further speculated that the two may have the same effect as targets and its mechanism.
