Effects Of Different Frequency Vibration Exercise On Breast Cancer Of Female Rats With Cancer-Related Fatigue

ObjectiveTo explore the change of cancer-related fatigue and the levels of factors IL-6,TNF-α and IL-1β in serum of female rats combined with different frequency vibration exercise by establishing the model of breast cancer-related fatigue in female Sprague-Dawley rats,in order to provide experimental basis for the treatment of breast cancer-related fatigue of exercise,while exploring IL-6 may be the important cytokines that involving in relieving cancer-related fatigue.Methods1.40 female rats were divided into two groups of normal control group(N,8 female rats) and breast cancer chemotherapy group,the breast cancer chemotherapy group were divided into four subgroups:model control group(C),low-frequency vibration exercise group(L),medium-frequency vibration exercise group(M),high-frequency vibration exercise group(H),8 female rats every group.For the breast cancer chemotherapy group,viable SHZ-88 breast cancer cells were injected into female rats,and were injected intraperitoneal epirubicin(2.5mg/kg/w) during six weeks after successful inoculation.The female rats of normal control group received same amount of saline.2.Exercise groups performed vibration exercise for 30(15min×2,5min intervals) minutes,the frequencies of three exercise groups were low frequency(15Hz),medium frequency(25Hz),high frequency(35Hz),the amplitude of the vibration was 1.5mm,1 time/day,6 day/week for 8 weeks.3.All female rats were anaesthetized after 8 weeks,drawn blood from abdominal aortal with anesthesia,through observed the general situations and the tail suspension test of these female rats to test cancer-related fatigue.The levels of factors IL-6,TNF-α and IL-1β were measured by ELISA.Results1.The general state shows:before the experiment,the difference of weight of female rats between different groups was not statistically significant(P>0.05).After chemotherapy,compared to N group,the weight of female rats in C group and three frequencies exercise groups was lower(P < 0.01).After exercise,compared to C group,the weight of female rats in L group and M group was higher(P<0.01),the weight of female rats in H group was not statistically significant(P>0.05).Compared to L group,the weight of female rats in M group was higher(P < 0.01),the weight of female rats in H group was lower(P <0.01).The weight of female rats in H group was lower(P<0.01) than that of female rats in M group.2.Rat tail suspension test:before the experiment,the difference of the immobility time of female rats between different groups was not statistically significant(P>0.05).After chemotherapy,compared to N group,the immobility time of female rats in C group and three frequencies exercise groups was longer(P < 0.01).After exercise,compared to C group,the immobility time of female rats in L group and M group was shorter(P<0.01),the immobility time of female rats in H group was not statistically significant(P>0.05).Compared to L group,the immobility time of female rats in M group is shorter(P<0.01),the immobility time of female rats in H group was longer(P<0.01).The immobility time of female rats in H group was longer(P<0.01) than that of female rats in M group.3.The result of cytokines shows:compared to N group,the levels of IL-6,TNF-α,IL-1β of C group were higher(P<0.01).Compared to C group,the level of IL-6 of three frequencies exercise groups was higher(P<0.01),the levels of TNF-α,IL-1β of L group and M group were lower(P<0.01),the levels of TNF-α,IL-1β of H group were not statistically significant(P>0.05).Compared to L group,the level of IL-6 of M group and H group was higher(P<0.01),the levels of TNF-α,IL-1β of M group were lower(P<0.01),the levels of TNF-α,IL-1β of H group were higher(P < 0.01).Compared to M group,the levels of IL-6,TNF-α,IL-1β of H group were higher,which the increase of the levels of TNF-α,IL-1β is statistically significant(P<0.01).ConclusionsThe weight loss and the tail immobility time prolong of female rats after inoculated SHZ-88 breast cancer cells and chemotherapy,the female rats appear fatigue and the female breast cancer-related fatigue model is established.After different frequency vibration exercise intervention,the low-frequency vibration exercise and the medium-frequency vibration exercise can relieve cancer-related fatigue,which especially the medium-frequency vibration exercise,while the high-frequency vibration exercise have no effect on cancer-related fatigue.The low-frequency vibration exercise and the medium-frequency vibration exercise can increase the level of factor IL-6,while reduce the levels of factors TNF-α and IL-1β,the cancer-related fatigue is significantly relieved,which especially the medium-frequency vibration exercise,the levels of factors TNF-α and IL-1β significantly increase after the high-frequency vibration exercise,which have no effect on the cancer-related fatigue.
