Compound Prunella Vulgaris is a nosocomial prescription which has good antihypertensive effect, and has been used for many years. Based on the theory of traditional Chinese Medicine, it is very important significance to study the antihypertensive site from Compound Prenella Vulgaris for the research and application of preparation, combined with modern production technology and methods.In our research, there are four major groups: process optimization, activity screening, chemical constituents and finger-print, for the antihypertensive site from Compound Prenella Vulgaris. In view of the aspect of actual production for pharmacyeutical companies, RSM(response surface method) was used to optimize extraction process, innovatively. Then an appropriate mathematical model was established to predict the best optimum condition.With macroporous adsorption resins, aqueous solution after precipitation from alcohol extraction were enriched and purified. Then 50% ethanol elution parts as the best active site was defined with the antihypertensive efficacy test. According to the organic spectroscopy technique, such as NMR, IR and MS, four monomeric compounds were isolated and identified from the best active part: compound I(rutin), compound II(5,6-dimethoxy-2-isopropenyl-benzofuran), compound III(5,7,4’-trihydroxy-dihydroflavonol) and compound IV(lupeol).By HPLC-DAD analysis method, finger-print of the best active site from Compound Prenella Vulgaris was established with calibration of the 26 common peaks, and the method showed high sensitivity and good reproducibility. It provides a reliable quality control method for the research of preparation.
