The research was on Xinjiang Morus nigra L.leaves as materials,and the results were as follows:1.Research on Nutrient Component and Active Ingredients of Mulberry Plant in Xinjiang Pro-vince,It was known that there are great differences in the contents of the components in different varieties of mulberry.The principal component analysis shows that the Morus nigra L.has a high-er value.2.Extraction condition of ultrasonic assisted extraction active components in mulberry leaf was optimized by the response surface method.The results showed that: when the extraction time at 67.75 min and 0.02 mol/L hydrochloric acid concentration,extraction temperature at 80,liquid materls ratio at 20.73 g/m L,volume fraction of ethanol 59.89%,and 3 times of extraction.Extraction rate: alkaloids 1.846±0.04 mg/g,polysaccharide 99.582±2.968 mg/g,flavonoids 12.901±0.474 mg/g.3.Analysizethat that the mulberry leaves in landing the hypoglycemic effect of material is polyhydroxylated alkaloids.The test on the content of total alkaloids and 1-deoxynojirinycin(1-DNJ),pigment content in Morus nigra L.leaves were determined,the results for total alkaloids in Morus nigra L.leaves are 2.8414 mg/g and 1-deoxynojirinycin(1-DNJ)content are 1.9414 mg/g;Two compounds in Morus nigra L.leaves were obtained analyzed by HPLC-MS/MS method as 1-deoxynojirinycin and fagomine.4.Through the different way for purification of the alkaloids,polysaccharides and flavonoids of alpha-glucosidase inhibitory activity were tested.The results showed that alkaloids and polysaccharides extract on alpha-glucosidase inhibitory activity significantly,and flavonoid extract on alpha-glucosidase inhibition activity was not obvious.The inhibitory activity of the compound was investigated.The results showed that the combination of alkaloids,polysaccharides and flavonoids was higher than the single action of flavonoids on injibitory activity.Coarse extract on alpha-glucosidase inhibitory activity inhibition mechanism is not clear,so the monomer compounds of alpha-glucosidase inhibition activity of test was designed.The results show that flavonoid glycoside of alpha-glucosidase inhibition activity is better.Through the monomer compound test,the result was that 1-deoxynojirinycin(1-DNJ)and flavone compounds after being used in combination showed good hypoglycemic effect,and the mechanism of decreasing blood sugar was not only.One of the mechanism was that by glycosidase inhibition,the decomposition and absorption of suger were inhibited,theother was that the insulin sensitivity was improved directly.After the merger of the composition the mechanism and advantages of decreasing blood glucose remained to be further study.
