| Objective: To detect and compare dynamic expression levels of miRNA146a、miR-4284、miR-4463、miR-4306、miR-210 and miR-221 in the peripheric blood circulation between patients with carotid artery stenosis and healthy control subjects,then explore the relationship among miRNAs and carotid artery stenosis and influence of intravascular stent implantation on miRNAs,and analyze the feasibility of those miRNAs to be biomarkers for carotid artery stenosis diagnosis.Methods: 27 patients with carotid artery stenosis who have had carotid stent implantation were chosen as case group,30 patients in hospital in the same period who had no carotid artery stenosis were chosen as control group.Elbow vein blood of two groups was extracted 2 days before surgery,within 24 h after surgery,postoperative 1 month and 3 months respectively.Total RNA was extracted from plasma,real-time quantitative PCR(RT-PCR)was used to detect dynamic expression quantity of miRNA146a、miR-4284、miR-4463、miR-4306、miR-210 and miR-221.SPSS13.0 statistical software was used to analyse data,numeration data was expressed as mean±standard deviation(x ±S),t test was used to compare means between groups,one-way ANOVA was used to compare multiple means in the groups,chi-square test was used to compare rate of sample.P < 0.05 was considered as statistically significant,each inspection was not less than three times.Results: Compared to the control group,(1)before carotid stent implantation,the expression level of miRNA146 a increased 5.48 times,miRNA4284 increased 13.83 times,miRNA4463 increased 23.37 times,miRNA4306 increased 21.02 times,miRNA210 increased 3.87 times and miRNA221 increased 3.05 times in the blood of patients with carotid stenosis,P< 0.05,(2)within 24 h after carotid stent implantation,the expression level of miRNA146 a increased to 14.17 times,miRNA4284 increased to 41.66 times,miRNA210 increased to 9.39 times,mi RNA221 increased to 19.07 times,while miRNA4463 and miRNA4306 dropped to 4.02 and 17.17 times respectively,showing statistically significant difference between groups(P < 0.05),(3)one month after carotid stent implantation,the expression level of miRNA146 a reduced to 11.26 times of control group,mi RNA4284 reduced to 6.17 times,miRNA4463 reduced to 2.5 times,miRNA4306 reduced to 13.39 times and miRNA210 reduced to 3.03 times of control group respectively(P < 0.05),while miR-221 expression level showed no significant difference between groups(P >0.05),(4)3 months after carotid stent implantation,the expression level of miRNA146 a upswinged to 12.85 times,miRNA4284 upswinged to 27.00 times,miRNA4463 upswinged to 108.47 times,miRNA4306 upswinged to 35.20 times and mi RNA210 upswinged to 24.48 times of control group respectively,all showed statistically significance(P < 0.05).In the comparison of before and after carotid artery stenting(1)plasma in patients with carotid stenosis,the expression of miR-221 significantly increased(P<0.05).When compared miRNAs expression level in patients with carotid artery stenosis between preoperative and postoperative,(1)miR-221 increased obviously(P< 0.05),the expression level atpreoperative,within 24 h after surgery,1 month after surgery and 3 months after surgery were all statistically significant(P < 0.05),(2)the expression level of miR-210 at preoperative,within 24 h after surgery and 3 months after surgery were statistically significant(P< 0.05),but the expression level at preoperative and 1 month after surgery showed no statistical significance(P > 0.05),(3)miR-4284 in 3months after surgery compared with preoperative,postoperative and 1 month after surgery,the expression level were all statistically significant(P< 0.05),(4)when miR-4306 in one month after surgery compared with 3months after surgery,the expression level was statistically significant(P < 0.05),while the other –s showed no statistical significance(P >0.05),(5)miR-4463 expression level in 3months after surgery was statistically significant(P < 0.05),the others were no statistical significance(P > 0.05),(6)miR146a-5 P expression level showed no statistical significance(P > 0.05). |
