Millettia speciosa,a kind of tropical medicinal plant,is widely distributed in Guangdong,Guangxi and Hainan Province.In this dissertation,the nutrient components,chemical components,extraction technology of flavonoids and quality control of Millettia speciosa in Hainan Province were investigated and the results were as follows:The nutrient components of Millettia speciosa in Hainan Province were assayed by means of ultroviolet spectrophotometry,high performance liquid chromatography(HPLC)and inductively coupled plasma mass spectrometry(ICP-MES).The results showed that the content of water,carbohydrates,protein,ash,fiber,oil was 9.27%,25.19%,5.44%,4.10%,32.02%and 1.57%,respectively.There were abundant mineral elements in Millettia speciosa and the content of Ca,Mg and Fe proved to be high.In addition,the content of heavy metal elements(Pb,As,Hg,Cu)was below the natianol limited standard.As for vitamins,the content of VB6 and VC proved to be high among eight kinds of vitamins.And there were 18 kinds of amino acids in Millettia speciosa and the essential amino acids accounted for 9.51%,which was deemed as superior protein resource according to EAAI estitament.Overall,Millettia speciosa was rich in nutrients,which was potential for new food resource’s development.On the part of chemical components,preliminary tests on the chemical components of Millettia speciosa in Hainan Province were conducted firstly.Then,the chemical components in Millettia speciosa in Hainan Province were separated with the methods of thin layer chromatography(TLC),silica gel column chromatography(SGCC)and HPLC and analyzed by GC-MS,1H-NMR and 13C-NMR.The lipid-soluble extract of ethyl acetate was analyzed by GC-MS,in which there were 45 kinds of chemical components identified accounting for 99.13%of the total.Ultimately,the chemical components in the extracts of ethyl acetate and n-butyl alcohol were separated and identified.There were five compounds identified,which were determined as indole-3-carboxylic acid,maackiain,4,4’-diacetoxy-1,1 ’-biphenyl,gancaonol and calycosin,respectively.Therein,indole-3-carboxylic acid and 4,4’-diacet--oxy-1,1’-biphenyl were separated from Millettia speciosa at first time.Based on single-factor experiments with.flavonoids extraction from the root ofMillettia Speciosa Champ.,response surface methodology was able to optimize the extraction process of flavonoids in Millettia speciosa in Hainan Province.The results attested that the model was good in fitting degree,little in error and good in repeatability.Then,the optimal condition of flavonoids extraction obtained according to regression model,significance analysis and actual situation was follows:the volume fraction of ethanol 80%,temperature 40 ℃,ultrasonic power 500 W and extraction times 3.The verification experiments were conducted in dupilicate for 6 times under this optimal condition,concluding that the average extraction rate was 1.31%and RSD was 1.56%.On the part of quality control,Millettia speciosa was extracted with ultra sonic based on the condition optimized with response surface methodology at first.Then,HPLC assays for maackiain and formononetin were established to determine the content of maackiain and formononetin,which feasibilities,simualtaneously,were verified by methodology expedition.Then,the content of water,ash,extracts,carbohydrates,heavy metal elements,pesticide residues and microbiological indexes were measured.According to the experimental results,the content of water was determined less than 10.16%,the content of ash was determined less than 5.01%and the content of the extract extracted by cold water,heated water,cold ethanol and heated ethanol was determined less than 5.54%,14.85%,1.08%and 2.44%,respectively while the content of carbohydrates,maackiain and formononetin was determined above 23.44%,3.04 μg·g-1 and 46.37 g·g-1,respectively.As for heavy metal elements,all samples met the demands of the limited stardard except that the content of Cd in the sample WN20140925 slightly exceeded the standard.In addition,for pesticide residues and microbiological indexes,the results showed all samples met the demands of the limited stardards.
