It is well known that excessive amount of free radicals in our body can cause cell injury and death,which may contribute to many diseases such as liver damage,aging,cancer,stroke,myocardial infarction,and so on.Pyran Chalcone compounds found in legume,moraceae plant have many kinds of biological activity,such as antioxidant,antibacterial,anticancer and antivirus.A series of pyran chalcone derivatives were synthesized in our laboratory previously.In this study,the anti-oxidative activities of these new chalcone derivatives were evaluated by cell oxidative damage model and chemical evaluation method.The cellular antioxidant mechanism and preventive effects of YZH-A087-26 on L-02 cell oxidation damage induced by carbon tetrachloride(CCl4)were also studied.Firstly,the antioxidant activities of 10 Pyran Chalcone compounds were evaluation by PC-12 cells hydrogen peroxide oxidation damage stress model and chemical method.Among them,compound YZH-A087-26 displayed the most potent anti-oxidative activity.Cell viability were increased significantly in YZH-A087-26-treated cells(89.91%),higher than the cell viability in DMSO-treated cells(22.03%)followed H2O2's treating.Moreover,YZH-A087-26's total reducing power of VC equivalent is 648.44 mg/mM,and the ABTS+radical scavenging capacity(EC50)is 0.54 mM.Secondly,we studied the effects of YZH-A087-26 on the LDH,GSH and DNA damage markers 8 hydroxy deoxyguanosine(8-OHdG)content in H2O2-treated PC-12 cells.The results showed that in YZH-A087-26-treated cells,cellular antioxidant enzymes GSH content is relatively higher,the LDH leakage will be lesser and the content of 8-OHdG will be relatively reduced than that in control cells.Finally,to verify the effectiveness of the compound YZH-A087-26 antioxidant activity in the liver,carbon tetrachloride(CCl4)stimulate normal liver cell(L-02)in acute liver injury study was performed.Following the treatment of CCl4,the survival rate of L-02 cells decreased to 42.19%,while 200 ?M YZH-A087-26 can improve survival rates to 81.60%.The results suggested that YZH-A087-26 has certain protective effect for the liver injury induced by CCl4.In conclusion,this study screened out YZH-A087-26 which has a higher antioxidant activity.It has significant protective effect of the cell oxidative damage induced by H2O2,and also has a good total reducing power and the ability to remove ABTS+ radical.In addition,it has certain protective effect for the liver injury induced by CCl4.
