The Research Of Antiviral And Antibacterial Activity Of Zhike Chuanbei Pipa Pill

ObjectiveIn clinic,Zhike Chuanbei Pipa Pill as an important Chinese medicine is often used to treat cough by cold and pharyngitis.Our study aims to assess anti-viral(Influenza virus FM1 strain,FM1,Herpes simplex virus type I,HSV-I,)and anti-bacterial(Staphylococcus aureus,alpha hemolytic streptococcus,S.pneumoniae,K.pneumoniae)activity of Zhike Chuanbei Pipa Pill.Methods1.To study the anti-influenza virus FM1 effects of Zhike Chuanbei Pipa Pill in mice.Sixty mice were randomly divided into 6 groups with 10 mice in each group:control group,influenza virus FM1 infected group,ribavirin treated group(60 times higher than the dose commonly used in clinics),Zhike Chuanbei Pipa Pill high dose group(30 times higher than the dose commonly used in clinics),Zhike Chuanbei Pipa Pill medium dose group(20 times higher than the dose commonly used in clinics),Zhike Chuanbei Pipa Pill low dose group(10 times higher than the dose commonly used in clinics).Mice were anesthesized with diethyl ether.The normal control group was given 50ul RPMI-1640 medium intranasally and the rest groups were infected with 4LD50 virus in 50ul volume intranasally.The normal control and virus-infected groups were given normal saline.The treatment group was given Zhike Chuanbei Pipa Pill and ribavirin by intragastric gavage at 0.4 ml per mouse for six days.The mice were given normal diet and drinking water.The protective effects of Zhike Chuanbei Pipa Pill on mice infected with influenza virus FM1 were studied by observing behavior changes and death rates.Lung Index,pulmonary pathological changes and the pulmonary virus hemagglutination titer were also used to assess the effects of Zhike Chuanbei Pipa Pill.The effects on immune system were studied using T and B lymphocyte proliferation test,spleen index and Enzyme Linked Immunosorbent Assay(ELISA).2.To study the anti-HSV-I effects of Zhike Chuanbei Pipa Pill in vitro.The anti-HSV-I effects of Zhike Chuanbei Pipa Pill were tested in Vero cells by the CPE and MTT methods.Rat sera containing the drug were prepared and their anti-HSV-I effects were also tested by the CPE and MTT methods.3.To study the anti-bacterial effects of Zhike Chuanbei Pipa Pill.Staphylococcus aureus,alpha hemolytic streptococcus,S.pneumoniae and K.pneumoniae were cultured on blood agar plates.Minimal inhibitory concentration(MIC)was determined using serial dilution method and drug susceptibility by paper diffusion tests.Results1 Mice were observed for their clinical response,growth and situation.Zhike Chuanbei Pipa Pill could significantly prolong the living time(P<0.01).The results of lung index,pulmonary pathological changes and the pulmonary virus hemagglitination titer proved that Zhike Chuanbei Pipa Pill had anti-inflammatory effect on pneumonia.The results of spleen index and spleen T and B lymphocyte transformation experiments proved that Zhike Chuanbei Pipa Pill could promote the immunological cells proliferate and strengthening the function on immune system.The TNF-a in srea increased after infected influenza virus.The TNF-a level decreased treated with zhike chuanbei pipa pill compared to model group(P<0.05).These show that Zhike Chuanbei Pipa Pill can inhibit TNF-a expression and reduce the inflammatory response.The IFN-? level increased compared to model group(P<0.05).This implied Zhike Chuanbei Pipa Pill can enhance the body's early anti-virus ability.2 HSV-I was inactivated directly by Zhike Chuanbei Pipa Pill.Absorption of HSV-I was inhibited obviously and the biological synthesis was intercepted.The best inhibitory rates were 75.41%,60.76%and 56.71%,respectively(P<0.05).The inhibitory rate of HSV-? increased with the raise of concentration of drugs.Serum with Zhike Chuanbei Pipa Pill had the anti-virus effect and the inhibition depended on the concentration and effect time.3 Each test tube show turbidity.Zhike Chuanbei Pipa Pill have no significant antibacterial circle against staphylococcus aureus,alpha hemolytic streptococcus,S.pneumoniae,K.pneumoniaeConclusion1 Zhike Chuanbei Pipa Pill has antiviral effects on influenza virus FM1in vivo2 Zhike Chuanbei Pipa Pill can strengthen function on immune system.3 Zhike Chuanbei Pipa Pill has the effects of anti-HSV-? in vitro.4 Zhike Chuanbei Pipa Pill has not significant effect on bactiorals in vitro.
