Lactate is not only an anaerobic fermentation product,or is the ‘culprit’ which could cause the body acidosis.In fact,the lactate could integrate and regulate multiple metabolic pathways as a regulatory molecule.Lactate takes part in the metabolic pathways of glycolysis,biosynthesis and biological oxidation as an important metabolic intermediate in spite of the lactate has no redox itself.The effect of lactate in the area of signal conditioning is reflected in the regulation of inflammatory cytokines,anti-inflammatory cytokines and the enzyme activity of central metabolic pathways.We study on lactate metabolic flux and control analysis of human in basal metabolic state motion state,learning state,fever state.The main results are presented.The metabolite change rate,metabolite concentration change and the enzyme activity of the serum which were isolated from the blood of volunteers under four different states were measured.The Fluoroskan Ascent FL was used to detect the enzyme activity based on the principle of enzyme coupling.The analysis of metabolic flux is a meathead which specialized measured and analyzed the flow of metabolic pathways,and is an important tool to calculate the metabolic flux in different pathways or in different conditions.The results indicated that the pyruvate dehydrogenase,phosphfructokinase,enolase,pyruvate kinase and citrate synthase had relatively higher positive regulatory effects on optimal steady state through the analysis of lactate flux control and the method of principal component analysis.And the effects of pyruvate dehydrogenase(CJPDHC= 0.246693),enolase(CJENO=0.28504)and citrate synthase(CJCS= 0.164945)were most obvious,which accounting for almost70% of the sun of flux control.Meanwhile the results of the flux control analysis also indicated that four enzymes had the negative regulation effect(CJLDH=-0.22416,CJα-KGDHC =-0.02533,CJG-6-PD=-0.27384,CJMDH=-0.21134).The lactate flux maps of human under different physiological states were draw and the metabolic flux ananlysis was took in this study.By studying on the flux control analysis of lactic acid salt during people in different physiological condition,we determined several enzymes which play an important role inthe metabolic flux of lactic acid salt.Because of lactic acid salt and cancer have close relationship,so we can from which we can screening theinhibitors of the key role of enzyme in lactic acid salt flux for cancer treatmen,or for the the development of health food products.
