Objective: To explore the intervention effect of Pingchuanning mechanism of cold asthma rats lung tissue in PI3 K signal transduction pathway in the treatment of cold asthma. Through the PI3 K signal transduction pathway observed in asthma rat s induced by factor PDGF、PI3K、PKB、NF-κB and other related gene, inve stigate the expression indexes about asthma related airway remodeling of TGF-β1and modulated effects of Pingchuanning, discuss the effect mechanism of Pingch uan ning inhibit airway remodeling in cold asthma rats.Methods: Totally 140 male SD rats were randomized into 7 groups,20 in each g roup.To copy the ovalbumin sensitized in rat asthma model.To sensitize rats by using 1ml suspension with intraperitoneal injection of 10% egg protein,aluminum hydroxide, physiological saline at the first,eighth days.At the fifteenth day, sed1% o valbumin solution ult RASonic atomization and gave the cold stimulation for 7 cons ecutive days,30 min enery time,one time every day.Modeled after 21 days,normal,model control groups were given the sanme amount of distillde water every day.the dexamethasone group,guilongkechuanning group and high、medium、low dose groups of Ping Chuan Ning decoction were given each drug drench according to h uman and rat body surface area than the conversion operator.After 4 weeks disse ctiong the rats and taking out the lung organization. the left lung with formalin fo r HE staining to observe the expression of preservation, the lung tissue pathological changes and immunohistochemical method to detect TGF-β1 in lung tissue; right lung into-80 C refrigerator preservation,used for RT-PCR PI3 K signal pathway in duced lung tissue factor PDGF mouse detection,signal transduction pathway of PDG F、PI3K、PKB、NF-κB.Results: Through HE staining can be found,compared with the normal group, mod el grouphad obvious airway remodeling phenomenon; compared with the model gro up, the treatment groups were properly alleviate airway remodeling. By immunohist ochemical method was compared with the normal group,the expression of PDGF、TGF-β1has the higher,the difference was statistically significant(P<0.01,P<0.05); a nd compared with the model group,Pingchuanning thetreatment group PDGF、TG F-β1 expression was decreased properly(P<0.01,P<0.05),especially in high and middle dose group decreased the Pingchuanning obviously, the best effect. Expressi on of by detecting the PI3 K signal transduction pathway of RT-PCR by PI3K、PKB、NF-κBm RNA that compared with the normal group,expressed in PI3K、PKB、NF-κBm RNA in asthmatic model increased(P<0.01,P<0.05); compared with model group,small dose group in Pingchuanning in the down-regulation of N F-κBm RNA expression is not obvious,the expressed in other groups, PI3K、P KB、NF-κBm RNA(P<0.01, P<0.05) have a appropriate declines.Conclusion:Pingchuanning can decrease asthma inducing factor PDGF、PI3K、P KB、NF-κB and airway remodeling factor TGF-β1 and can alleviate the airway inflammation of asthmatic rats through regulate PI3 K signaling pathway, and can effectively reverse the airway remodeling.
