Objective:The observe the effects of electro-acupuncture,enriched rehabilitation training and electro-acupuncture combined with enriched rehabilitation training on neural function and nerve regeneration inhibitory factor Myelin-associated glycoprotein(MAG) and Oligodendrocyte-Myelin glycoprotein(OMgp) protein expression at different time points in rats with cerebral ischemia reperfusion injury.From the view point of regulating central nerve regeneration inhibitory factor, the mechanism of electro-acupuncture combined with enriched rehabilitation training on promoting the recovery of nerve function after cerebral ischemic was discussed, which provided a new experimental basis for cerebral ischemia.Methods:1. 24 rats of the sham operated group were taken out randomly from 120 SPF Sprague-Dawley rats.The rest of the rats were used to establish focal middle cerebral artery occlusion reperfusion model by using the right external carotid artery ligation method,and randomly divided into the model group,electro-acupuncture group, enriched rehabilitation group, electro-acupuncture combined with enriched rehabilitation group.Each group was divided into three subgroups of 3d,7d and14 d,and there was 8 rats in every group. Electro acupuncture therapy set dilatational wave,5~100 times/s, 20min/days. The enriched rehabilitation therapy contained the enriched environment and the balance beam,rotating bar,net screen,drum training,10 min/days per item.The enriched environment was changed every 3 days.2. Neurological deficits and sensation,motor function score of each group were evaluated by the method of nerve function rehabilitation;the pathological changes in hippocampus CA1 area of rats were observed by HE(hematoxylin and eosin staining)staining;the expression of MAG and OMgp positive cells in hippocampus CA1 area ofrats in each group were analyzed by immunohistochemistry technique;the protein expression of MAG and OMgp were observed by Western Blot detection technique.Results:1. HE stainingUnder the optical microscope, the neurons in the hippocampus CA1 area of the sham operation group were stained uniformity and morphological structure was complete;in the model group,the edema neurons were obvious and arranged in sparse;in hippocampus CA1 area of the electro-acupuncture group and enriched rehabilitation group,neuron tumefaction was reduced;in the electro-acupuncture combined with enriched rehabilitation group,swelling of nerve cells was significantly reduced,the morphological structure tended to be normal.2.Marks of neurological function and sensation, motor functionAfter electro-acupuncture treatment,the balance beam training, rotating bar training,screen,each score comparison found that the neurological function in the sham operation group was normal.Before treatment,compared the model group with the three treatment groups, there was no statistically significant difference(P>0.05);At 3d,compared with the model group,the scores of the three treatment groups had decreased,but there were no statistical significance(P>0.05),scores between the three treatment groups were not statistically significant(P>0.05);At 7d,14 d,the scores of the three treatment groups were decreased.Compared with the model group score,the scores of the electro acupuncture group and enriched rehabilitation group were lower trend obviously(P<0.05),the difference of electro acupuncture combined with enriched rehabilitation group had statistical significance(P<0.01).Scores between the three treatment groups were compared, the differences of scores between the electro-acupuncture group and enriched rehabilitation group were not statisticalsignificance(P>0.05),at 7d,the score of the electro-acupuncture combined with enriched rehabilitation group was lower than that of the enriched rehabilitation group and the electro acupuncture group,there were statistically significant difference(P<0.05);at14d,the score of the electro-acupuncture combined with enriched rehabilitation group is lower than that of the electro-acupuncture group and enriched rehabilitation group, the difference is significant(P<0.01).2.Expression of MAG, OMgp proteinAt 3d, 7d, 14 d after cerebral ischemia, the hippocampus CA1 of rats were detected by immunohistochemical staining and Western blot technique. Through statistical analysis,MAG and OMgp protein in hippocampus tissue of the sham operation group showed a basic expression,and the expression of MAG,OMgp protein in the hippocampus CA1 of the remaining four groups rats were high expression. MAG,OMgp protein expression of the model group was higher than that of the sham operation group, there were statistically significance(P<0.01).At 3d after surgery,The expressions of MAG,OMgp in the four groups were all enhanced,and there were no significant differences in the expression of the three treatment groups(P>0.05);At 7d,the expression of MAG,OMgp in the four groups reached the peak,and had a downward trend at 14 d,but it was still higher than the postoperative 3d.At 7d,14 d,compared with the model group,MAG,OMgp protein expressions of the electro acupuncture group and enriched rehabilitation group were decreased,there were statistically significant(P<0.05),differences of the electro-acupuncture combined with enriched rehabilitation group were statistically significant(P<0.01).A comparison between the three treatment groups, the differences of scores between the electro-acupuncture group and enriched rehabilitation group were not statistical significance(P>0.05);at 7d,the MAG,OMgp protein expression of electro acupuncture combined with enriched rehabilitation group was lower than that of the enriched rehabilitation group and the electro-acupunct-ure group,there were statistically significant(P<0.05),at14 d,compared the electro-acupuncture combined with enriched rehabilitation group with the electro-acupuncture group and rehabilitation group, the differences were statistically significant(P<0.01).Conclusions:1. After cerebral ischemia reperfusion injury,therapy of electro-acupuncture,enriched rehabilitation training and electro-acupuncture combined with enriched rehabilitation can improve hippocampus pathology damage and promote the recovery of neural function defect,and the electro-acupuncture combined with rehabilitation therapy was superior to the simple therapy of electro-acupuncture and enriched rehabilitation training;2. The electro-acupuncture combined with enriched rehabilitation can promote the recovery of nerve function and the mechanism may be related to inhibiting the expression of OMgp and MAG protein.
