objective:Studies on the Chemical constituents in fresh tubers of Dioscorea Bulbifera L.and determination of the content of diosbulbin B placed in different years of Dioscorea Bulbifera.to reasonable use of resources.Method:Dioscorea Bulbifera.refluxing and extraction with 95% ethanol,extraction by petroleum ether, ethyl acetate, n-butanol.Different parts was separated by Silica gel column chromatography, gel chromatography on Sephadex LH-20 column and ODS;The use of mass spectrometry, and nuclear magnetic resonance spectroscopy of the monomer were isolated structure determination.The Gemini-C18(250 mm×4.60 mm,5 μm)column was used. The mobile phases consisted of methanol-0.1% phosphoric acid water(35: 65), the flow rate was 1.0m L·min-1.the detection wavelength was 210 nm and the column temperature was 25℃.Result:A total of 18 compounds were isolated as Diosgenin(1), stigmasterol(2), 3,7-dimethoxy-5,3’,4’-trihydroxy flavone(3), 2, 7-dihydroxy-3,4-dimethoxyphenanthrene(4), 3,7-dihydroxy-2,4-dimethoxy phenanthrene(5), 2-Hydroxy-7,10,10-trimethoxy-anthracen-9-one(6), 2,7-dihydroxy-4-dimethoxyphenanthrene(7), 2,7-dihydroxy-4-methoxy-9, 10-dihydroxy phenanthrene(8), nonanedioic acid(9), 8-epidiosbulbin E acetate(10), 1, 7-bis-(4-hydroxy phenyl)-4E,6E-heptadien-3-one(11),Diosbulbin B(12),pentacosanoic acid 2′, 3′-dihydroxypropyl ester(13), 2, 7-dihydroxy-4-methoxy-9,10-dihydroxy-phenanthrene(14), 1, 7-bis-(4-hydroxyphenyl)-hepta-1E, 4E, 6E-heptatrien-3-one(15), 6-ethoxy-1H-pyrimidine-2, 4-dione(16), 3, 5, 4’-trihydroxy- bibenzyl(17),Diosbulbin F(18).The linear concentration range of diosbulbin B was at 5.36-53.60 μg·m L-1(R=0.9996). The RSD was 3.37%.Conclusion: 18 compounds were isolated from the plant, 4 compounds were first isolated from the plant,which 2-hydroxy-7,10,10-trimethoxy-10H-anthracen-9-one was a new compound.The content of diosbulbin B placed in different years was similar.The content of diosbulbin B in stem was little,it can be ignored.The content of diosbulbin B in the leaves was about 1/5 of the content of tuber and can be effectively used.
