Objective: In recent years, A number of studies have indicated that mi RNA plays an important role in the occurrence and development of gastric cancer,the research of micor RNA(mi RNA) and gastric cancer ha s become one of the hot spots in medical research. At present, the relat ionship between mi RNA-185 expression in gastric cancer and lymph no de metastasis with tumor cell apoptosis is not clear.In this study, we det ected the expression of mi RNA-185, cell apoptosis rate and the expressi on level of four kinds of apoptosis protein in gastric cancer and lymph node metastasis.We also make a preliminary study on the above proble ms by analyzing the relationship between them and their significance,wh ich provides a basis for further understanding of the mechanism of mi R NA-185 in gastric cancer.Methods: We collected gastric cancer tissues and metastatic lymph node tissues of fresh specimens from October 2014 to July 2015 during the surgical treatment of Patients with advanced gastric cancer in fourt h hospital of Hebei Medical University, a total of 46 cases.(1).Take the U6 gene as a benchmark, use real-time fluorescence quantitative PCR(q RT-PCR) to detect the level of mi RNA-185 expression of the 46 cases, and then statistical analysis.(2).The apoptosis rate of gastric cancer tissue s and metastatic lymph nodes were detected by flow cytometry, and the relationship between the expression of mi RNA-185 and the cell apopto sis rate was analyzed.(3).using immunohistochemical method to detect Th e expression of Bcl-2, Bax, Survivin and Fas protein in gastric cancer t issues, metastatic lymph node tissues.Using SPSS19.0 statistical analysis software for statistical description and analysis of relevant data, which was statistically significant differences P < 0.05.Results: 1 The expression of mi RNA-185 increased compared with lymph node metastasis in gastric cancer tissues(t=5.813, P=0.000), the difference wa s statistically significant.2 The apoptosis rate of gastric cancer was significantly higher than that of metastatic lymph node tissue apoptosis rate(8.49% + 1.59% vs 4.37% + 1.69%),(t=12.04, P=0.000) the difference was statistically signific ant.3 The rate of apoptosis were positive correlation between the expression level of mi RNA-185 in gastric cancer tissues and metastatic lymph node tissues(r=0.602, P < 0.001; r=0.514, P=0.001).4 Bcl-2, Bax, Survivin, Fas were expressed in gastric cancer tissues a nd metastatic lymph node tissues. The positive expression levels of Bcl-2 protein and Survivin proteinin metastatic lymph node tissues were hig her than that in gastric cancer tissues(x~2=4.563,P=0.033,x~2 =4.842 P=0.028), the difference was statistically significant.Bax protein,Fas protei n, contrary to the above results((x~2=5.718, P=0.033,x~2 =4.696,P=0.030), the difference was statistically significant.5 Spearman correlation analysis:5.1 In gastric carcinoma tissues,the expression level of Bax protein and Fas protein was positively correlated with the cell apoptosis rate(r=0.451, P < 0.01; r=0.409, P < 0.01),the expression level of Bcl-2 protein a nd Survivin protein was negatively correlated with the apoptosis rate(r=-0.551, P < 0.01, r=--0.814, P < 0.01).5.2 In metastatic lymph node tissues,the apoptosis rate was positively co rrelated with the level of expression of Bax protein(r=0.563, P < 0.01),the expression level of Bcl-2 protein and Survivin protein was negativel y correlated with the apoptosis rate(r=-0.389, P < 0.01, r=-0.643, P < 0.01).The expression level of Fas protein was not correlated with cell a poptosis P=0.616.5.3 In gastric carcinoma tissues,the expression levels of mi RNA-185 with that of Bcl-2 protein, Survivin protein were negatively correlated(r=-0.451, r=-0.872, P < 0.001, P < 0.01),that of Bax protein,and Fas protei n were no correlation(P=0.642, P=0.739).5.4 In metastatic lymph node tissues,the expression level of mi RNA-185 was positively correlated with Bax protein expression(r=0.398, P < 0.01),the expression level of mi RNA-185 was negatively correlated with B cl-2 protein and Survivin protein expression(r=-0.697, P < 0.01; r=-0.794, P < 0.01).Conclusion: 1 The expression of mi RNA-185 in lymph node metastasis was lo wer than that in gastric carcinoma,Suggesting that the expression of mi R NA-185 further decrease in the process of gastric cancer metastasis, and the expression of mi RNA-185 may be involved in the occurrence and metastasis of gastric carcinoma in some mechanism.2 The apoptosis rate was significantly lower than that in gastric ca ncer tissues and metastatic lymph node tissues, and the apoptosis rate w as positively correlated with the expression of mi RNA-185 in both tissu es.Suggesting that the ability of cell apoptosis decreased in the course o f gastric cancer metastasis,mi RNA-185 may regulate apoptosis to be inv olved in the occurrence and metastasis of tumor cells.3 Expression of Bcl-2, Bax, survivin, Fas in gastric carcinoma and metastatic lymph nodes were different.In gastric cancer, Bcl-2, Bax, Fas, survivin and tumor cell apoptosis rate are correlated.There was a correl ation between the expression of Bcl-2, Bax and Survivin in lymph node metastasis and tumor cell apoptosis, and the expression of Fas was not correlated with the rate of apoptosis.Indicates that Bcl-2, Bax, survivin and Fas is involved in the process of gastric cancer occurrence, Bcl-2, Bax, survivin is involved in the process of metastasis of gastric cancer and gastric cancer in the transfer process can not use primary tumor ap optosis protein expression situation instead of metastatic foci in the expr ession.4 The expression of mi RNA-185 and Bcl-2, survivin were correlate d in gastric cancer, The expression of mi RNA-185 and Bcl-2、Bax、Sur vivin were correlated In metastatic lymph nodes.Suggested that mi RNA-185 may affect apoptosis through regulating the expression of Bcl-2 and Survivin in gastric cancer, and participate in the occurrence of gastric cancer,and mi RNA-185 may affect apoptosis through regulating the expr ession of Bcl-2, Bax and Survivin in metastatic lymph node tissue, and participate in the transfer of gastric cancer.5 In summary,The decreased expression of mi RNA-185 induced apo ptosis rate decreased in gastric transfer process.Gene expression regulate d by mi RNA-185 is changing during the course of gastric cancer metast asis, and the regulation of cell apoptosis is also changing.
