Mongolian medicine is the important component section of traditional medicine, it has complete theoretical system and abundant treatment practice. Base on the long term practical experience, Mongolian medicine gathered a large variety of medicines, which were used to treat and prevent chronic and difficult diseases. Costing less money, it is effective, easy and green. Acnoitum Kusnezoffii Reichb, also called Caowu, is a Mongolian medicine commonly used as a analgesic to treat plague, strokes, rheumatism etc. But because its toxicity usually causes poisoning or dying, Terminalia chebula Retz (Hezi) processing was commonly adopted to reduce the toxicity of Caowu for clinical therapy. To confirm the mechanism of reducing toxicity, this study is designed to investigate the chemical changes of Hezi during processing. The main research contents and results are as follows:(1 Application of FTIR, we compare the IR spectra of different extracts and processing sample, the results shown that the water extract facilitate the dissolution of bioactive components in Hezi herb. The toxicity of Caowu decreases as the increase of Hezi processing time.(2)An simple and selective HPLC-DAD method for determination and quantification of gallic acid and ellagic acid was developed and validated. The method was used to determinate the content of these 2 compounds in different time and proportion processing sample. The result showed that the effect of reducing toxicity of Caowu is positive correlated to processing time and Hezi quantity.(3) Combined with the previous results, we used LC-MS to further detect Hezi single decoction and different Hezi-Caowu proportion processing sample,8 compounds including punicalagin,chebulic acid, gallic acid, corilagin, Tellimagrandin I(2,3-di-O-galloyl-4,6-(S)-HHDP-a/b-D-Glc),ferulic acid,chebulagic acid and ellagic acid.In conclusion, the study investigate the chemical basis of Hezi processing Caowu, with FTIR、HPLC and LC-MS. Results revealed that the variation of tannin content in was impact ed significantly by processing, it suggested that tannin of Hezi might plays a important role in reducing toxicity of Caowu. The scientific conclusion is the key to guide further mechanism re search and clinical medication. |
