Apoptosis, an important mechanism of organism to maintain the physiological balance, is a cell death pathway regulated by a series of genes. The apoptotic mechanism and pathway are a hot research realm rising in life science and clinical medicine since the concept was been proposed by Kerr, et al in 1972, and it has been being in the ascendant so for. After decades of endeavor, the role of apoptosis in the medical field is gradually uncovered. Apoptosis affects profoundly the cell proliferation, senility, canceration and the occurrence of many diseases, especially closely related to the tumor sensibility, metastase tendency and recurrence risk.The apoptotic dection has become more accurately with the rapid development of molecular biology and laboratory technology. Currently, the apoptosis detection method includes three types: morphology observation, immunohistochemical assay and biochemical assay. All these assays qualitatively or quantitatively present the change of characteristics during cell apoptosis, which is very helpful for diagnosis of diseases triggered by apoptosis and treatment of cancer.Based on the deficiency of existing methods and the basic knowledge of apoptosis, this paper proposes a new method called diffraction image quantification parameter analysis based on diffraction-imaging flow cytometry(DIFC) for research of apoptosis of HL-60 cell line. The main works are summarized as follows:With HL-60 cells synchronized through serum deprivation and induced by hydrogen peroxide(H2O2) as study samples, this paper firstly used tradional flow cytometry to analyze the apoptosis rate at different post-treatment times. Secondly, fluorescence images were carried on the statistic analysis of apoptosis rate and two-dimension morphometric data assisted by Hoechst 33342、Annexin V-FITC/PI. Afterwards, this paper acquired a number of diffraction image of side scattering light utilizing DIFC and applied gray-level co-occurrence matrix(GLCM) to extract 34 feature parameters of diffraction images that have been preprocessed to analyze the correlation of feature parameters and apoptosis rate. Combing with the results of three detective methods, this paper calculated their coefficient of determination to evaluate the significance of quantitative diffraction image analysis in cancer cell study, and proposed potential directions for further researches.
