| Objective:To test the expression level of multidrug resistant proteins P- gp, MRP1, and GST-π before and after neoadjuvant chemotherapy in oscc,and then observe their clinical significances. Methods:1. We have collected 77 cases of oral squamous carcinoma pathological specimens with complete clinical data from the First Affiliated Hospital of Nanchang University from 2000-2011(including 50 valid groups and 27 invalid group).The envision immunohistochemical has been adopted to determine expression level of P-gp, MRP1, and GST-π proteins.Furthmore, the 3 and 5 year survival rates have been compared.2. We have collected 18 OSCC patients as valid groups and 15 OSCC patients as invalid group from the oral and maxillofacial surgery department of First Affiliated Hospital of Nanchang University since 2013. Western Blot has been employed to detect the expression level of P-gp, MRP1, and GST-π proteins.3. We have collected 18 OSCC patients as valid groups and 15 OSCC patients as invalid group from the oral and maxillofacial surgery department of First Affiliated Hospital of Nanchang University since 2013, RT-qPCR has been employed to detect the expression level of P- gp, MRP1, and GST-π mRNA. Results:1. The histological type, clinical stage, and pathological grading between valid and invalid group had no statistical significance(P > 0.05), and the cases had comparability.2.The total effective rate of neoadjuvant chemotherapy in O SCC was 64.9%, the remission rate was 9.88%.3. The Envision Immunohistochemical suggested that positive expression rate of p-gp, MRP1, and GST-π before and after chemotherapy was rised both valid and invalid group, and had significant respectively(p < 0.05);The raise of P- gp, MRP1 positive expression rate between valid and invalid group was statistically significant(P < 0.05),but not GST-π(P > 0.05).4. The Envision Immunohistochemical also suggested that there was no relationship between gender, pathological grading, clinical stage and lymph node metastasis with the positive expression of P-gp, MRP1, and GST-π proteins in valid and invalid group before chemotherapy(P > 0.05).5. Western Blot and RT-qPCR suggested that expression of P-gp, MRP1, and GST-π had been enhanced in the valid and invalid group before and after chemotherapy,while the expression variation of P-gp, MRP1 showed significance between the valid and invalid group(P < 0.05).6. Kaplan- Meier survival analysis methods analyzed that the 50% survival rate of survival time of valid group was 5 years,while 2.5 years of invalid group; And the median survival time of valid group was longer than invalid group(p < 0.05), the 3and 5-year survival rate of valid group was better than invalid group(p < 0.05). Conclusion:The overexpression of P-gp and MRP1 were related to therapeutic outcome and long-term survival rate of neoadjuvant chemotherapy in OSCC; valid group showed superior 3,5 years survival rate. |
