Objective To investigate the therapic effects of NLXTC on brain ischemia-reperfusion injury in rat, neurological scores, r CBF, apoptosis, infarct size and ERS injury were explored the mechanism of action of NLXTC, as well as to observe whether MEK/ERK pathway is the molecular biology mechanism of healing ischemia-reperfusion injury.METHODS1.Protection effect of NLXTC on I/R inrats(1)Middle cerebral artery occlusion(MCAO) model was established by suture-occluded method. Garcia JH's score was used to measure neurological scores at the points of 24 h, 7d and 14 d after cerebral ischemia injury in rats.(2)The regional cerebral blood flow was monitored during the modeling, reperfusion after 24 hour of ischemia,7days after modeling and 14 days after modeling respectively.(3)Transmission Electron Microscope(TEM) was used to observe the impact of NLXTC on the ultrastructure of hippocampal neurons after cerebral ischemia damage in rats.(4) TUNEL stain was used to observe the impact of NLXTC on apoptosis of neuron.(5) Measured infarct area by TTC staining2.Nneuroprotection by Naoluoxintong capsules is mediated through inhibition of endoplasmic reticulum stress(1)Western Blotting method was used to detect the effect of NLXTC on SBDP145expression(2) Western Blotting method was used to detect the effect of NLXTC on TRPC6expression(3)Immunofluorescence technique was used to detect the effect of NLXTC on TRPC6expression(4)Western Blotting method was used to detect the effect of NLXTC on GRP78,?GRP78,CHOP,Caspase-12,p-CREB expressionRESULTS1.Protection effect of NLXTC on I/R inrats(1) NLXTC evidently decrease neurological scores of Brain I/R rats at 7d and 14 d after cerebral ischemia injury in rats. It manifests that NLXTC have effects to reduce the injury of nervous system.(2) The r CBF was restored in the NLXTC at 7d and 14 d after cerebral ischemia injury in rats. It indicate that NLXTC can improve neurological action by increasing r CBF,which play a protective effect on cerebral ischemia-reperfusion injury.(3) TEM images showed that the complete cell morphology was visible in sham group.Compared with model group,NLXTC administration group organelle damage morphology is improved significantly,prominent nuclei could observe obviously.NLXTC could remakably improve the damage of neurons.(4) NLXTC administration can significantly inhibit I/R induced apoptosis at 14 d after cerebral ischemia injury in rats.(5) NLXTC administration can significantly inhibit Infarct volume2.Nneuroprotection by Naoluoxintong capsules is mediated through inhibition of endoplasmic reticulum stress NLXTC blocks a II-spectrin breakdown product--SBDP145 formation(1) NLXTC can decrease the level of SBDP145(2) NLXTC inhibit the degradation of TRPC6, improved the TRPC6 levels induced by cerebral ischemia reperfusion injury.(3) Immunofluorescence technique shows that NLXTC capsules increased the cerebral ischemia reperfusion model rats TRPC6 expression.(4) NLXTC activate after CREB, promote the expression of p- after CREB, improve the neuron survival ability.(5)NLXTC increase the expression of GRP78, promote the endoplasmic reticulum in its protection.(6) NLXTC can inhibit apoptosis induced by cerebral ischemia reperfusion injury signal molecule expression of CHOP and caspase- 12..CONCLUSIONS1. The method of middle cerebral artery occlusion was a successful way to establish cerebral ischemia injury in rats.2. NLXTC has an significantly effect on the injury induced by cerebral ischemia 2H reperfusion injury in rats.3.NLXTC can significantly improve the ER stress mediated ischemia reperfusion injury...
