Hericium erinaceus is a kind of fungus of Hericium erinaceus(Rull ex F.) Pers. It contains various biologically active substances, such as protein, vitamins, mineral elements and other nutrients. H. erinaceus is a common edible and medicinal fungus. It has the function of enhancing immunity, anti-aging and anti-oxidant activities(Seoung Rak Leea et al., 2015; Dilani et al., 2012).The extracellular polysaccharides(EPS) and intracellular polysaccharides(IPS) were extracted through the method of water extraction and alcohol sedimentation. The EPS and IPS were purified, separated and their monosaccharide composition was evaluated. By the analysis of GC, the extracellular polysaccharides(EPS) were composed of arabinose, mannose, galactose and glucose with a ratio of 1:7:14:52, and the composition of intracellular polysaccharides(IPS) was rhamnose, xylose, mannose, galactose and glucose with a ratio of 3:4:7:14:137.This article analyzed the molecular link and the functional groups of EPS-1、EPS-2、IPS-1and IPS-2.The molecular link of EPS-1 includes N-H、O-H、C=C、C=N、N=N、saturated C-H bond and unsaturated C-H bond. The functional groups includes hydroxyketone complex、monomer acid、acid complex、amino、imido、alkene group、azo、CH3、CH2 and aromatic groups.The molecular link of EPS-2 includes N-H、O-H、C=C、C=N、N=N. The functional groups includes monomer acid、amino、imido、alkene and azo.The molecular link of IPS-1 includes N-H、O-H、C=C、C=N、N=N、saturated C-H bond and unsaturated C-H bond. The functional groups includes hydroxyketone monomer and its complex、monomer acid、amino、imido、alkene group、oxime、azo、CH3、CH2、aromatic groups and alkyno.The molecular link of IPS-2 includes N-H、O-H、C=C、C=N、N=N. The functional groups includes amino、alkene、azo.The study makes a preliminary validation of H. erinaceus polysaccharide’s in vitro antioxidant capacity and take the DPPH, hydroxyl radical and total reducing power as an index. Experiments show that both the IPS and EPS have a very good scavenging effect on the above-mentioned radicals. The IPS and EPS have potential antioxidant capacity.The protective effects of the IPS and EPS from H. erinaceus on the CCl4-induced hepatic injury of mice were investigated in this work. The model of hepatic injury of mice was induced by CCl4 and three tested levels(200, 400, 800 mg/kg) of EPS and IPS were set as the experimental group. Results showed that the aspartate aminotransferase(AST) and glutamic pyruvic transaminase(GPT) activities in serum were reduced by the supplement of EPS and IPS, while the blood lipid levels(CHOL, TG and ALB) were improved. In liver tissue, the lipid peroxidation(LPO) and malondialdehyde(MDA) were largely decreased, and the superoxide dismutase(SOD) and catalase(CAT) activities were significantly increased. The evidence demonstrated that the EPS and IPS of H. erinaceus were protective for liver injury. The histopathological observations of mice liver slices indicated that EPS and IPS had obvious effects on liver protection.
