Objective:To study the influence of simo decoction on mice intestinal bacterial diversity with constipation and then provide the basis for its curative effect mechanism.Methods: The 36 experimental mice randomly were divided into model group, normal group, Simo decoction and Zhi Zhu decoction treatment groups. Then a model of constipation was built. After the model was established, the small intestinal microbes total DNA of six normal group and model group mice were taken. The treatment groups respectively received Simo decoction and Zhi Zhu Decoction, while the model group and normal group were given sterile water by gavage for 6d, and then the small intestinal microbes total DNA was extracted for sequencing analysis of 16 s rDNA V4 area.Results: Through the building constipation,in the total number of OTU, model group had 896 different species, less than normal group of 982 species, 807 of them were the same; from the point of PCA analysis, normal group were relatively concentrated distribution, while the model group were relatively discrete distribution and the distance with the normal group were large; from the point of biological diversity index, Chao index, ACE index, Shannon index of model group were significantly lower than normal group(P < 0.01 or P < 0.05), model group Simpson index value were significantly higher than that of normal group(P < 0.05); from the point of relative abundance of bacteria,Coriobacteriales,Clostridia, Clostridiales, Deltaproteobacteria, Bacteroidales, Coriobacteriia, Desulfovibrionales relative abundance in model group were significantly lower than normal group(P < 0.01 or P < 0.05), while Actinobacteria, Bifidobacteriales, Erysipelotrichales relative abundance were significantly higher than that of normal group(P < 0.01). Treatment after constipation building spleen-deficient, Simo decoction and Zhi Zhu decoction had no significant effect on intestinal bacteria community diversity, the difference between them was the influence of the relative abundance on intestinal bacteria, specifically, compared with control group, the relative abundance of Erysipelotrichi, Allobaculum, Betaproteobacteria Erysipelotrichales, Burkholderiales, Erysipelotrichaceae, Sutterella were increased obviously(P< 0.05 or P< 0.01) and Firmicutes,Bacilli, Lactobacillales, Lactobacillaceae,Lactobacillus were reduced obviously(P< 0.05 or P< 0.01) in Zhi Zhu decoction group,while Simo decoction group had significantly reduced the relative abundance of Bacilli, Lactobacillales(P < 0.05); compared with model group, the relative abundance of Actinobacteria,Erysipelotrichi,Betaproteobacteria, Bifidobacteriales,Erysipelotrichales,Burkholderiales,Bifidobacteriaceae,Erysipelotricha ceae,Alcaligenaceae,Bifidobacterium,Allobaculum,Sutterella were increased obviously(P< 0.05 or P< 0.01) and Firmicutes,Bacilli,Clostridia,Lactobacillales,Clostridiales, Lactobacillaceae,Ruminococcaceae, Lactobacillus,Lachnospiraceae,Ruminococcus were reduced obviously(P< 0.05 or P< 0.01), while Simo decoction group had significantly reduced the relative abundance of Firmicutes,Bacilli,Lactobacillaceae, Lactobacillus,Ruminococcaceae, Ruminococcus(P< 0.05);compared with Zhi Zhu decoction group, Simo decoction group had significantly reduced the relative abundance of Burkholderiales, Alcaligenaceae, Sutterella(P< 0.05).Conclusion: The mice intestinal bacterial diversity with constipation had reduced obviously.Clostridia,Coriobacteriales,Bacteroidales etc were the important gut bacteria related to mice with constipation.Simo decoction can adjust the species composition of Firmicutes,Bacilli,Lactobacillale etc to achieve therapeutic effect on constipation,and Simo decoction can more significantly adjust intestinal bacterial diversity than Zhi Zhu decoction.
