The traditional Chinese medicine (TCM) is an important part of the natural plant resources. As a kind of potential antioxidant resources, TCM can scavenge the free radicals, restore oxidation material, restrain the delay or prevent oxygen free radicals which may cause some physical disease and food decaying effectively. Therefore they have been widely used in pharmaceutical and food, etc.The Huangqi (Astragalus membranaceus (Fisch.) Bge. var mongholicus (Bge.) Hsiao) (AME)-Baishao(Paeonia Lactiflora Pall, Paeoniaceae)(PL) herb pair is one of the most widely used herb pairs and has appeared in many traditional Chinese medicine formulas(TCMF)which have a good curative effect in remeding dysmenorrhoea and repairing spirit blood. Both the two herbs are existed in the traditional polyjuice potion Astragalus chienchung soup and Huangqi Guizhi Wuwu Decoction. In this paper, we studied the antioxidant activity, changes in the chemical components using GC-MS and LC-MS in different solvent extracts of AME when combined with PL. The mainly results were as follows:1. The vitro antioxidant assay (ABTS and DPPH free radical scavenging assay, ?-carotene bleach text) was applied to evaluate the antioxidant capacity of AME and PL herb pair. It was found that the antioxidant ability of AME and PL herb pair was significantly higher than that of single herbs. Besides, the content of polyphenol and flavonoid changed after AME was combined with PL, which may be one of the reasons bringing about synergistic antioxidant effect.2. The chemical components of AME and PL herb pair were detected using the GC-MS. It was found that there were 15components in the petrol ether extract of the herb pair. Most of these components are higher fatty acids and esterifications, especially riched in linoleic acid and flax acid methyl ester. In the other hand, we found components such as Paeonol, Eugenol, ?-cadinene from PL and Caryophyllene, ?-sitosterol, Lupenone from herb pair are all having antioxidant ability. The new components identified in herb pair explained there may be facilitation of dissolving components during the extract processing, or because of some chemical or physical changes.3. The oxidation resistance and the synergistic antioxidant effect of different samples extracts by ethyl acetate at different concentrations in soybean oil were also studied by determin. The result indicated that both the two single herb and herb pair were all have an inhibit effect to the oxidation of soybean oil. PL had the strongest inhibit effect. Besides, it was found that there was a concentration dependent of samples during a certain concentration range. Compaired with BHT used alone, single herb and herb pairs combined with isoconcentration BHT expressed a higher antioxidant capacity, which illustrated there may had a synergistic antioxidant effect between the extracts and BHT.4. In our study, we detected the chemical components of the ethyl acetate extracts of AME, PL and herb pair by HPLC. It was found that there were 28 components in the ethyl acetate extracts of the herb pair. Furthermore,16 constituents were identified by the HPLC-DAD-MS/MS technique, which mainly belong to flavoniod and terpenoid. Moreover, most of the flavoniods came from AME and most terpeniods came from PL. Such components as calycosin, isocarthamidin, onion and lactiflorin, paeoniflorin, hydroxyl-paeoniflorin are important active ingredients from AME and PL respectively, which have potential antioxidant properties.
