Objective:Laryngeal squamous cell carcinoma(LSCC)is one of the common malignant tumors in head and neck, although with the continuous progress of diagnostic techniques, the treatment of laryngeal cancer has taken a variety of surgery combined with chemotherapy drugs and new radiation therapy, the efficacy has been significantly improved.The occurrence of laryngeal cancer is produced by a variety of factors, there are many studies on laryngeal cancer, but the molecular mechanism of its occurrence and development is still not completely understood. Therefore, the study of the role of signaling pathway in laryngeal cancer is very necessary.To study the relationship between the expression of large tumor suppressor homolog 2(LATS2), Merlin protein, Transcriptional coactivator with PDZ- binding motif(TAZ) in laryngeal squamous cell carcinoma and adjacent normal tissue, the relationship between laryngeal squamous cell carcinoma and other tumors and the relationship between them and the mechanism, which provides a certain theoretical and experimental basis for the study of laryngeal cancer with specific therapeutic targets.Methods: 38 cases pathology specimens of laryngeal squamous cell carcinoma patients as the experimental group selected from attendance in otolarygology department of the second Hospital of Hebei Medical University in June 2014 to September 2015 while collecting the 38 cases adjacent tissue from tumor 0.5cm as control group. All specimens were no history of radiation and chemotherapy before surgical excision,and preoperative biopsy pathological examination were identified as laryngeal squamous cell carcinoma.Immunohistochemistry SP method was used to detect the expression of LATS2, Merlin, TAZ in laryngeal squamous cell carcinoma tissue and adjacent tissue,and its relationship with the clinicopathological parameters, as well as the expression of LATS2, Merlin, TAZ and the correlation between the three.All the experimental data were statistically analyzed by SPSS13.0, and test to P < 0.05 as the difference was statistically significant.Results:1 Expression of LATS2Expression of LATS2 in laryngeal squamous cell carcinoma and adjacent tissues:The positive expression rate of LATS2 in laryngeal carcinoma tissue was 26.32% lower than in the adjacent tissues that the positive expression rate was 65.79%, and the difference between the two groups was statistically significant(P < 0.05).The relationship between LATS2 expression and clinicopathological parameters in laryngeal carcinoma:The positive expression rate of LATS2 in highly differentiated squamous cell carcinoma was 81.82% significantly higher than in the middle and low differentiated laryngeal squamous cell carcinoma that the positive expression rate was 3.70%, the difference between the two groups was statistically significant(P < 0.05);the positive expression rate of LATS2 in laryngeal squamous cell carcinoma without lymph node metastasis was 42.86% significantly higher than in laryngeal squamous cell carcinoma with lymph node metastasis that the positive expression rate was 5.88%, the difference between the two groups was statistically significant(P < 0.05).While LATS2 in age ≤ 60 years old patients with positive expression rate of laryngeal squamous cell carcinoma was 23.53%,in age > 60 years old patients with positive expression of laryngeal squamous cell carcinoma rate was 28.57%, the difference between the two groups was not statistically significant(P > 0.05);LATS2 in smoking less than 20 cigarettes per day in patients with positive expression rate of laryngeal squamous cell carcinoma was 25.00%, the positive expression rate of laryngeal squamous cell carcinoma was 26.92% in patients with smoking more than 20 cigarettes per day,the difference between the two groups was no statistical significance(P > 0.05);The positive expression rate of LATS2 in the glottis laryngeal squamous cell carcinoma was 28.57%,and the positive expression rate was 20% in the upper and lower glottis laryngeal squamous cell carcinoma,the difference between the two groups was not statistically significant(P > 0.05);The positive expression rate of laryngeal squamous cell carcinoma in UICC stage I ~ II period was 35%,The positive expression rate of laryngeal squamous cell carcinoma was 16.67% in the period of III ~ Ⅳ,the difference between the two groups was not statistically significant(P > 0.05).2 Expression of MerlinExpression of Merlin in laryngeal squamous cell carcinoma and adjacent tissues:The positive expression rate of Merlin in laryngeal carcinoma tissue was 39.47% lower than in the adjacent tissues that the positive expression rate was 81.58%, and the difference between the two groups was statistically significant(P < 0.05).The relationship between Merlin expression and clinicopathological parameters in laryngeal carcinoma:The positive expression rate of Merlin in highly differentiated squamous cell carcinoma was 81.82% significantly higher than in the middle and low differentiated laryngeal squamous cell carcinoma that the positive expression rate was 22.22%, the difference between the two groups was statistically significant(P < 0.05);The positive expression rate of Merlin in laryngeal squamous cell carcinoma in UICC stage I ~ II period was 65.00% significantly higher than the positive expression rate of laryngeal squamous cell carcinoma in the period of III ~ Ⅳ was 11.11%,the difference between the two groups was statistically significant(P < 0.05);the positive expression rate of Merlin in laryngeal squamous cell carcinoma without lymph node metastasis was 57.14% significantly higher than in laryngeal squamous cell carcinoma with lymph node metastasis that the positive expression rate was 17.65%, the difference between the two groups was statistically significant(P < 0.05).While Merlin in age ≤ 60 years old patients with positive expression rate of laryngeal squamous cell carcinoma was 49.06%,in age > 60 years old patients with positive expression of laryngeal squamous cell carcinoma rate was 33.33%, the difference between the two groups was not statistically significant(P > 0.05);LATS2 in smoking less than 20 cigarettes per day in patients with positive expression rate of laryngeal squamous cell carcinoma was 41.67%, the positive expression rate of laryngeal squamous cell carcinoma was 38.46% in patients with smoking more than 20 cigarettes per day,the difference between the two groups was no statistical significance(P > 0.05);The positive expression rate of Merlin in the glottis laryngeal squamous cell carcinoma was 39.28%,and the positive expression rate was 40.00% in the upper and lower glottis laryngeal squamous cell carcinoma,the difference between the two groups was not statistically significant(P > 0.05).3 Expression of TAZExpression of TAZ in laryngeal squamous cell carcinoma and adjacent tissues:The positive expression rate of Merlin in laryngeal carcinoma tissue was 76.32% higher than in the adjacent tissues that the positive expression rate was 13.16%, and the difference between the two groups was statistically significant(P < 0.05).The relationship between TAZ expression and clinicopathological parameters in laryngeal carcinoma:The positive expression rate of TAZ in highly differentiated squamous cell carcinoma was 36.36% significantly lower than in the middle and low differentiated laryngeal squamous cell carcinoma that the positive expression rate was 92.59%, the difference between the two groups was statistically significant(P < 0.05);The positive expression rate of TAZ in laryngeal squamous cell carcinoma in UICC stage I ~ II period was 60.00% significantly lower than the positive expression rate of laryngeal squamous cell carcinoma in the period of III ~ Ⅳ was 94.44%,the difference between the two groups was statistically significant(P < 0.05);the positive expression rate of TAZ in laryngeal squamous cell carcinoma without lymph node metastasis was 61.90% significantly lower than in laryngeal squamous cell carcinoma with lymph node metastasis that the positive expression rate was 94.12%, the difference between the two groups was statistically significant(P < 0.05);The positive expression rate of TAZ in the glottis laryngeal squamous cell carcinoma was 92.86% significantly higher than the positive expression rate was 30.00% in the upper and lower glottis laryngeal squamous cell carcinoma,the difference between the two groups was statistically significant(P < 0.05).While Merlin in age ≤ 60 years old patients with positive expression rate of laryngeal squamous cell carcinoma was 76.47%,in age > 60 years old patients with positive expression of laryngeal squamous cell carcinoma rate was 76.19%, the difference between the two groups was not statistically significant(P > 0.05);LATS2 in smoking less than 20 cigarettes per day in patients with positive expression rate of laryngeal squamous cell carcinoma was 83.33%, the positive expression rate of laryngeal squamous cell carcinoma was 73.08% in patients with smoking more than 20 cigarettes per day,the difference between the two groups was no statistical significance(P> 0.05).Conclusions:1 The positive expression rate of LATS2 and Merlin in laryngeal squamous cell carcinoma was lower than the tissues adjacent to carcinoma, while the positive expression rate of TAZ in laryngeal squamous cell carcinoma was higher than the tissues adjacent to carcinoma;The expression of LATS2 only associated with pathological grading and presence of lymph nodes, and it was not associated with age, smoking amount, tumor clinical stage and the location of tumor; the expression of Merlin was related to the clinical stage, pathological grading and presence of lymph nodes, and it was not related to the age, the amount of smoking and the location of tumor; The expression of TAZ was related to clinical stage, pathological grade, location of tumor and presence of lymph nodes, and it was not related to the age, the amount of smoking.2 LATS2, Merlin, TAZ has correlation among the three. Expression level of LATS2 and Merlin was positively correlation, which may suggest that the two may play a synergistic role in the occurrence and development of laryngeal cancer; TAZ was highly expressed in laryngeal squamous cell carcinoma and negatively correlated with Merlin and LATS2, which may suggest that TAZ may provide some direction for the diagnosis of laryngeal cancer, targeted therapy and prognosis.And the three are related to the Hippo signaling pathway.
