Cancer has been one of the most dangerous diseases with increasing morbidity and mortality rate.Early detection of malignant tumors and their metastasis is a vital factor for good patients’ prognosis.It has been found that GLUTs are over-expressed in human cancers.As a cancer imaging strategy the Warburg effect has garnered a great deal of interest in recent years.Some methods are clinically used,18FDG-PET e.g.However the methods in existence always have some drawbacks such as radiation exposure and low image quality.Herein the paper was focused on the design of NIR florescent DCPO glucose analogues which may make for early detection of tumors.The structure of NIR florescent dye DCPO was modified in this paper.With a better fluorescence property,N3-DCPO was improved from DCPO to connect with glucose.The distance between florescent and glucose is adjusted by the length of polyethylene glycol.Five probes containing glucose were prepared,we proceeded to dye cancer cells.The paper selected MCF-7、MKN-45、HeLa as the three cancer cell lines.After the processes of western blots and inhabitation experiments,we proved that the uptake of compounds was via GLUT-1.The best distance between DCPO and glucose was found out,and N2 could be the best in the probes prepared.Synthesized several probes containing glucose and DCPO,we confirmed the best length of pacer arm and the most effective analogue.The new compound can be used for developing the early detection of malignant tumors.Meanwhile the new dye DCPO can also be applied to screening of new drugs,because of its drug-like structure and remarkable fluorescence properties.
