The Expression Of Tumor Metastasis Associated Protein 2(MTA2) In Bladder Cancer And The Effect Of MTA2 On The Biological Characteristics Of Bladder Cancer Cells | | Posted on:2017-10-14 | Degree:Master | Type:Thesis | | Country:China | Candidate:K N Peng | Full Text:PDF | | GTID:2334330485973374 | Subject:Immunology | | Abstract/Summary: | PDF Full Text Request | | Objective: To investigate the expression of tumor metastasis associated protein 2(MTA2)in bladder cancer and to explore the effect of MTA2 on the biological characteristics of bladder cancer cells.Methods:1 The expression levels of MTA2 in bladder cancer tissues and non-cancer tissues were detected by Immunohistochemical method,and the correlation between MTA2 protein expression or clinicopathological characteristics of patients were also analyzed.2 The expression levels of MTA2 in bladder cancer cells were detected by Western blotting method,including T24,EJ,J82 et al.3 The expression levels of MTA2 in bladder cancer cells with MTA2 over-expression or low-expression were detected by RT-PCR and Western blotting method.4 The proliferation,migration and invasion abilities of bladder cancer cells with MTA2 over-expression were detected by MTS,Clone formation,Wound healing,transwell migration and matrigel invasion chamber assays,respectively.5 The proliferation,migration and invasion abilities of bladder cancer cells with MTA2 low-expression were detected by MTS,Wound healing,transwell migration and matrigel invasion chamber assays,respectively.Results:1 Immunohistochemical method demonstrated that the expression levels of MTA2 in bladder cancer tissues were higher than those in the non-cancer tissues(P<0.01).The expression level of MTA2 was associated with clinical stage,histological grade,invasion into lymph and distant metastasis(P<0.05),not age/year,gender and tumor size(P>0.05).2 Western blotting method demonstrated that the expression levels of MTA2 in T24 were lower than EJ and J82(P<0.05),but the expression levels of MTA2 in EJ were higher than T24 and J82(P<0.05).3 RT-PCR and Western blotting method demonstrated that the expression levels of MTA2 in T24 with MTA2 over-expression were higher than CD511 B and no-treatment T24,and the expression levels of MTA2 in EJ with MTA2 low-expression were lower than NC and no-treatment EJ.4 MTS assay demonstrated that over-expression of MTA2 promoted proliferation abilities of T24(P<0.05),while low-expression of MTA2 inhibited proliferation abilities of EJ(P<0.05).5 Clone formation assays demonstrated that over-expression of MTA2 promoted clone formation abilities of T24(P<0.01).6 Transwell migration assays demonstrated that over-expression of MTA2 promoted migration abilities of T24(P<0.01),while low-expression of MTA2 inhibited migration abilities of EJ(P<0.01).Matrigel invasion chamber assays demonstrated that over-expression of MTA2 promoted invasion abilities of T24(P<0.01),while low-expression of MTA2 inhibited invasion abilities of EJ(P<0.01).7 Wound healing assays demonstrated that over-expression of MTA2 promoted Wound healing abilities of T24(P<0.01),while low-expression of MTA2 inhibited Wound healing abilities of EJ(P<0.01).Conclusions:The expression level of MTA2 in bladder cancer tissues were higher than those in the non-cancer tissues.The expression level of MTA2 was associated with clinical stage,histological grade,invasion into lymph and distant metastasis,not age/year,gender and tumor size.MTA2 promoted proliferation,migration and invasion abilities of bladder cancer cells. | | Keywords/Search Tags: | bladder cancer, MTA2, metastasis, proliferation, migration, invasion | PDF Full Text Request | Related items |
