This dissertation is composed of three chapters. The first chapter summarized the progress of purification and bioactivity of polysaccharides. The second chapter studied the antithrombotic mechanism of polysaccharides extracted from Rubus spp. Blackberry seeds. The third chapter studied the procagulant activity and tyrosinase activity of polysaccharides from Angelica dahurice roots.Chapter 1. Advances of purification and bioactivity of polysaccharidesThe progress of purification and bioactivity of polysaccharides were summarized.Chapter 2. Antithrombotic mechanism of polysaccharides in Rubus spp. Blackberry seedsThe crude polysaccharides were obtained from the seeds of Rubus spp. Blackberry. Sevage reagent and dialysis bag were used to remove proteins and small molecular weight impurity, respectively. Then, BSP-1a, BSP-1b, BSP-2 and BSP-3 were isolated by anion exchange chromatography and sephadex column chromatography.The results of anticoagulant activity of BSP-1a, BSP-1b, BSP-2 and BSP-3 indicated that BSP-1b, BSP-2 and BSP-3 exhibited the anticoagulant activity in vitro. Therefore, the anti-thrombosis effects of BSP-1b, BSP-2 and BSP-3 were evaluated. The results suggested that BSP-1b, BSP-2 and BSP-3 had the inhibition effect on thrombus formation in blood stasis model rats, and the antithrombotic activities were associated with the regulation of vascular endothelium active substance, activating blood flow and anticoagulation activity.Chapter 3. Procoagulant and tyrosinase inhibitory activity of Angelica dahurice roots polysaccharidesFive polysaccharides ADPs-1a, ADPs-1b, ADPs-2, ADPs-3a and ADPs-3b were isolated from Angelica dahurice roots by DEAE-cellulose-52 followed with Sephadex G-100 column chromatography. The coagulant activities of the polysaccharides were evaluated by activated partial thromboplastin time(APTT), thrombin time(TT), prothrombin time(PT) and fibrinogen(FIB) in vitro. Coagulation assays indicated that ADPs-1b had procoagulant activities and exerted the procoagulant activities through intrinsic pathway, extrinsic pathway and increased the content of FIB in vitro. ADPs-2 had procoagulant activities and exerted the procoagulant activities through intrinsic pathway and extrinsic pathway. ADPs-3a had procoagulant activities and the activity was associated with the intrinsic pathway and increased the content of FIB. ADPs-3b had procoagulant activities and exerted the activities through extrinsic pathway and increased the content of FIB. The activity of tyrosinase was detected of polysaccharides from A. dahurice. All polysaccharides had inhibitory effect against tyrosinase in vitro.
