Objective On the basis of former single factor research, through to non small cell lung cancer of ERCC1, RRM1 expression and EGFR mutation research, find between may potential co expression or reverse expression relationship, for patients with chemotherapy and target to treatment and medication chosen to provide theoretical basis.Method Application branched DNA liquid phase chip technology after the sample cracking, Pre hybridization, repeated washing, hybridization, the RNA capture, signal amplification and detection, Luminex assay to determine mRNA expression level of detection and threshold. According to the value of the detection of 150 samples, using median statistical methods to determine the expression of each gene threshold (the one with cutoff value), and according to the pre calculated sensitivity, specificity, accuracy, positive predictive value, negative predictive value, threshold assessment reliability; according to one with cutoff value, according to the high or low expression of division calculated expression and distribution proportion. Application of Spearman rank correlation test of relativity, the basic relationship between gene expression and clinical features. Application of Wilcoxon rank sum test to compare the relationship between the expression of EGFR, RRM1 and ERCC1.Result Age was negatively correlated with ERCC1:the older the age, the lower the expression of ERCC1, the pathologic type was positively correlated with RRMl(r=-0.222, p<0.01). The expression level of RRM1 in patients with squamous cell carcinoma was higher than that in patients with adenocarcinoma(r=-0.267, p<0.01). Pathological type was negatively correlated with EGFR:the expression level of EGFR in patients with squamous cell carcinoma was lower than that in patients with adenocarcinoma(r=-0.258, p<0.01). T staging was positively correlated with RRM1:the higher the grade of T, the higher the level of RRM1 expression(r=0.179, p<0.05). N staging was positively correlated with RRM1:the higher the N stage, the higher the level of RRM1 expression (r=0.199, p<0.05). The degree of differentiation was negatively correlated with RRM1:the higher the degree of differentiation, the lower the level of RRM1 expression(r=-0.204, p<0.05). Smoking history was negatively associated with ERCC1:the level of ERCC1 expression in smokers was lower than that in patients without smoking(r=-0.178, p<0.05).ERCC1,RRM1 and expression were negatively correlated with the expression of EGFR(r=-0.214,p<0.05;r=-0.240,p<0.01), the level of ERCC1 expression was positively correlated with the expression of RRM1 (r=0.264, p<0.01)ConclusionFor NSCLC, ERCC1 gene expression level is correlated withpatient age and smoking history; RRM1 gene expression is correlated with clinical stage, histological type and differentiation.The curative effect and prognosis of ERCC1 in SCLC expression and the platinum drugs showed negative correlation;In NSCLC cells, the expression levels of the three genes showed a marked feature of co expression. According to the results of this study, patients with different pathological types of RRM1 and EGFR expression exists negative correlation; ERCC1 expression and patient age and history of smoking correlated; RRM1 expression and clinical staging, pathological types and degree of differentiation were correlated. The clinical data of NSCLC patients may be used as a reference index to predict the efficacy and survival time of chemotherapy drugs in patients with clinical data.
