| Objective:Through the method of injection of Complete Freund’s Adjuvant(CFA)into the prostate,the rat model of chronic nonbacterial prostatitis(CNP)is established.To explore the expression changes of the glial fibrillary acidic protein(GFAP)in the dorsal horn of spinal cord after intrathecal injection of the brain natriuretic peptide(B-type natriuretic peptides,BNP)and to investigate the impact of intrathecal injection of BNP on the chronic pain caused by chronic nonbacterial prostatitis.Method:1.150 Healthy male SPF SD rats were randomly divided into CNP+ intrathecal injection of physiological saline group(control group),CNP+ intrathecal injection of BNP group(BNP)group,intraprostatic injection of physiological saline + intrathecal injection of physiological saline(sham operation group),and each group 50 rats.A stable model of the CNP was established at the 7 days after intraprostatic injection of CFA.2.BNP group was Intrathecal injection of BNP,the other groups were intrathecal injection of physiological saline,L6-S1 spinal dorsal horns were extracted after the continuous injections for 1d、2d、3d、4d、5d.3.To detect the expression changes of GFAP in the spinal dorsal horn after the continuous intrathecal injection into each rat with the technology of real-time quantitative PCR and Western Blot.Result:1.Real-time quantitative PCR results showed that:the expression of GFAP in the control group was upregulated compared to the sham operation group(p<0.05).Compared to the control group,the expression of GFAP was decreased after the continuous intrathecal injection of 1d、2d、3d、4d、5d(p<0.05).Then 1d,2D,3D,4D,5D were 0.84±0.07、0.50±0.04、0.54±0.03、0.67±0.04、0.53±0.01 times.2.After the continuous intrathecal injection of 1d、2d、3d、4d、5d,the protein expression of GFAP in the control group increased significantly compared to sham operation group(p<0.05).and the protein expression of GFAP wan significantly decreased compared to the control group BNP(p<0.05).Conclusion:Intraprostatic injection of CFA in rats can cause the upregulating expression of GFAP in L6-S1 spinal dorsal horn.Continuous intrathecal injection of BNP can induce the downregulating expression of GFAP in L6-S1 spinal dorsal horn of the CNP model rats.But the level of reduction has no significant correlation with the continuous intrathecal injection days.Intrathecal injection of BNP may inhibit the chronic pain induced by CNP. |
