Background: The incidence of coronary heart disease caused by atherosclerosis increased year by year,which is seriously harmful to life and health.The related experiments in our group showed that there was significant correlation between some factors and AS in the course of the occurrence and progress of AS,such as PAPP-A,LOX-1,IL-1β,MMP-9,MMP-12 and TIMP-1 et al.Fibronectin(Fn)and its receptor integrin alpha 5 beta 1(integrin α5β1)are highly concerned with the migration and proliferation of various malignant tumor cells.There are not analysis and report on the expression of local integrin α5β1 and Fn in human coronary atherosclerotic plaque.Purpose: We detected the expression of CD68,actin,integrin α5β1,Fn in aorta and coronary artery tissues as well as in the atherosclerotic plaque,analysised the function and correlation between Fn and integrin α5β1 in occurrence and development of atherosclerosis.Method: In this study,we used immunohistochemical staining method to detect the expression of CD68,actin,Fn and integrin α5β1 in aorta and coronary artery.The degree of coronary artery stenosis was determined by elastic fiber staining and NIH Scion Image(60.1)software,and the dyeing of CD68 and actin were used to assist the localization in the atherosclerotic plaque.According to the different stages of atherosclerotic plaque,the tissues were divided into two groups: the fat pattern and atherosclerotic plaque group.The coronary artery tissues were divided into A group(< 25%),B group(25%-50%),C group(50%-75%)and D group(>75%)according to the degree of stenosis.According to whether there was a history of acute myocardial infarction,the cases were divided into ACS group,old myocardial infarction group and normal group.Result:1.The expression of integrin α5β1 in the smooth muscle cells inner elastic plate was stronger than in the tunica,the results were same in the aortic group(P=0.003)and coronary group(P=0.000).2.In aortic,the fatty streaks group expression of integrin α5β1 in smooth muscle proximal elastic plate was stronger than atherosclerotic plaque group(P=0.000).3.There was no significant difference in the expression of integrin α5β1 in tunica smooth muscle cells between the aorta and the coronary artery with atherosclerosis(P=0.227),but in two group the expression of integrin α5β1 higher than control group(P= 0.008,P=0.012).4.The expression of integrin α5β1 in the smooth muscle cells showed the significant difference between A group and C group(P=0.000);A group and D group(P=0.000);B group and C group(P=0.075);B group and D group(P=0.001),but A group and B group(P=0.510);C group and D group(P=0.520).5.The high expression region of focal calcification integrin α5β1 was stronger than the non-calcified in the coronary atherosclerotic plaque(P=0.000).6.There was no significant difference in the expression of integrin α5β1 in smooth muscle cells in ACS group,old myocardial infarction group and control group(P=0.754,0.870,0.805).7.Expression of Fn in aorta: The expression of Fn in the inner elastic lamina was higher than in the tunica(P=0.000).The expression of Fn in the inner elastic lamina of the inner membrane in the fatty streaks group was stronger than the atherosclerotic plaque group(P=0.008).8.The expression of Fn in the endometrial stromal cells was stronger than medial smooth muscle stroma of the coronary artery atherosclerosis group(P=0.000),and Fn and Integrinα5β1 had a certain positive correlation(R=0.902,P=0.000).Conclusion: 1.Integrin α5β1 was involved in the regulation of smooth muscle cell migration,even the occurrence and development of atherosclerosis.2.Synergistic effect of Fn and integrin α5β1 promotes occurrence and development of atherosclerosis.3.The expression of integrin α5β1 in the coronary artery showed a tendency to decrease with the increase of the degree of coronary artery stenosis.4.There was no significant correlation between the expression of integrin α5β1 in coronary artery and the occurrence of acute coronary syndrome.
