| A large number of previous studies have confirmed that the pharmacological effects of Cordyceps polysaccharide having anti-tumor, antioxidant, immunomodulatory, liver and kidney protection. In this study, immunology, biology-related principles and techniques can be used to study the immunomodulatory effects of artificial fermented Cordyceps powder and its polysaccharides from the angles of non-specific immunity and specific immunity, cellular immunity and humoral immunity, cytokines and antibodies. And try to explore its antioxidant activity. Provide theoretical support for the pharmacological effects of cordyceps and its deep-processed products. The main contents of this paper are as follows:We chose the mouse peritoneal macrophages as target cells to discuss immune function of Cordyceps powder polysaccharide and purified polysaccharide on macrophages in vitro. The results showed that it could significantly enhanced peritoneal macrophage proliferation activity and phagocytic function, it could also improve the cytokine secretion of TNF-α, IL-1β,IL-6.We chose spleen lymphocytes in immunosuppressed mice as target cells to explore influence of Cordyceps powder polysaccharide and purified polysaccharide on spleen lymphocyte immune function in vitro. It was found that both polysaccharides could enhance Con A induced T lymphocyte and LPS induced B lymphocyte proliferation, in addition, T lymphocyte subsets CD4+ / CD8+ ratio was improved obviously, and so that cyclophosphamide induced immunosuppression got some recovery, the polysaccharides could also significantly increase the secretion of IL-4, IL-6, TNF-α and IL-1 β in spleen lymphocyte supernatant, suggesting that Cordyceps polysaccharide could regulate immune function.We constructed immunosuppressive mice model to study the immune regulation and antioxidation effects of fermented Cordyceps powder and its polysaccharide in vivo. The results showed that the fermented Cordyceps powder and its polysaccharide could effectively enhance Con A induced T lymphocyte and LPS induced B lymphocyte proliferation, T lymphocyte subsets CD4+ / CD8+ ratio was improved significantly, with the increase of fermented Cordyceps powder and its polysaccharide dose, IL-6, IL-10 and TNF-α in splenic lymphocyte supernatant, as well as Ig M in serum were increased, and activities of SOD and CAT were also improved, MDA content decreased. However, there were no significant changes in Ig M and Ig A in the supernatant of splenic lymphocytes. These showed that the fermented Cordyceps powder and its polysaccharide could effectively inhibit the immunosuppressive effect of cyclophosphamide, and could improve the antioxidant ability of the immunosuppressive mice.In summary, the fermented Cordyceps powder and its polysaccharide could play a role in immune regulation through affecting peritoneal macrophages and splenic lymphocytes, and they also had antioxidant effects in vivo, mechanisms of immune regulation and antioxidation require in-depth study. |
