| Purpose: The anti human hepatoma cells of Pseudolaric acid B in vitro(HepG2)mechanism was studied systematically.Method:Cell counting and cell morphology methods were used to determine the growth of Pseudolaric acid B cells;influence of the scratch test corter Pseudolaricis percutaneous acetic acid effect on metastasis of hepatocellular carcinoma(HCC)by flow cytometry detection corter Pseudolaricis percutaneous acetic acid on the cell cycle;fluorescence staining method was applicted to detect the effect of Pseudolaric acid B treatment after HepG2 cell tubulin aggregation;real time PCR was served as discovering the effects of Pseudolaricis percutaneous acetic acid on transcription of p53 gene;Western blot was used to detect the cell associated protein expression.Result: Cell counting method and morphological observation confirmed Pseudolaric acid time dependently inhibit growth of HepG2 cells,and the morphology and expression of proteins related to cell apoptosis was induced by Pseudolaric acid B;Pseudolaric acid treatment can effectively inhibit cell transfer;analysis of the cell cycle found corter Pseudolaricis percutaneous acetic acid blocked HepG2 cells in G2/M phase detection;fluorescence staining confirmed Pseudolaric acid can promote cytoskeletal protein aggregation;detection of transcription of P53 gene showed that Pseudolaric acid reduced the transcription of P53 gene;protein expression showed that the expression of Pseudolaric acid P53 protein,P27,regulation of pro-casepase-3,BaX,Bcl2,FAS and FASL protein.Conclusion: Pseudolaric acid B on in vitro culture of HepG2 cell growth significantly inhibited,and through the distribution of microtubules in control cells to inhibit cell transfer and regulation of tumor cell cycle and apoptosis related proteins induced cell apoptosis to the antitumor effect of,also found that p53 protein and p27 protein in the anti tumor effect of Pseudolaric acid B. |
