MethodsImmunocytochemistry,Western Blot and Real time-quantitative polymerase chain reaction (qRT-PCR) were used to analyze the change of the quantity of protein and gene expression at day 0,days 10,days 20 and day 30 during cell culture of hESC directional differentiation to retinal photoreceptor progenitor cells.ResultsThe HI cell was long spindle and net-like gathered,it can be gathered into a ball after grown in embryoid bodies medium 1 days later, embryoid bodies can attached to the Matrigel on the first day,around the cell mass gradually had climbed out, the cell morphology changed from the original class round to long spindle, forming a rosette shape about day 10 and the axon elongation increased gradually, similar to nerve cells.Otx2 expression is always positive In the process of retinal photoreceptor progenitor cells, ssea4 and Ki67 expression gradually reduce, Sox2 expressed positively.ConclusionsImproved SFEB/DLFA differentiation method can make the embryonic stem cells differentiationed to retinal photoreceptor progenitor cells efficiently without serum and feeder.Thedifferentiation of retinal photoreceptor progenitor cells maintained a strong split proliferation, has part of the stem cell characteristics, while the cells gradually mature, important markers of RPC of the retinal photoreceptor cells such as Otx2 and Sox2 increased, but have not differentiated to mature photoreceptor cells.
