Objective:Observation of electro-acupuncture on rats with type II diabetic peripheral neuralgia pain behavior and spinal cord p-JAK2 and p-STAT3 expression of,explore the mechanism of electro-acupuncture in treatment of diabetic neuropathic pain.Rats regularly bundled control group and model groupMethods:Fourty male Sprague-Dawley rats aging 6 week,8 randomly selected as control group.The remaining 32 rats prepared with type 2 diabetes peripheral neuropathy model according to the literature method and molded 24.Model rats were randomly divided into three groups(n=8):model group,electropuncture group and acupuncture group.Control group and model group were regularly tied up,electropuncture group and acupuncture group select "jump ring" and "Yanglingquan" treatment for 3 weeks.On day 0(T0),day 56(T1),day 70(T2)measurement of fasting glucose,insulin concentration and mechanical paw withdrawal response threshold(PWT),in the first96 days(T3)measured only PWT.Upon completion of all measurements T3 in taking chloral hydrate anesthesia lumbar spinal cord was observed by Western blotting assessment of spinal p-JAK2 p-STAT3 expression.Result:(1)The difference in weight of rats in each group was not statistically significant at T0(P>0.05);Weight was significantly higher in model group,electropuncture group and acupuncture group at T1,the difference was statistically significant(P < 0.05),Weight was significantly higher in model group,electropuncture group and acupuncture group at T2,the difference was statistically significant(P <0.05).(2)Blood glucose level of rats in each group no significant difference at T0(P>0.05);Blood glucose level of rats in each group no significant difference at T1(P> 0.05);Blood glucose level was significantly higher in model group,electropuncture group and acupuncture group,the difference was statistically significant at T2(P<0.05).(3)Differences insulin concentrations and insulin sensitivity coefficient of the rats no significant difference at T0(P>0.05);model group,electropuncture group and acupuncture group insulin concentrations and insulin sensitivity index was significantly higher,the difference was statistically significant at T1(P<0.05);model group,electropuncture group and acupuncture group insulin concentrations and insulin sensitivity index was significantly higher,the difference was statistically significant at T2(P<0.05).(4)PWT value was no significant difference between the groups of rats at T0(P>0.05);PWT values still no statistical significance of the rats(P>0.05);PWT value of the control group was significantly higher than model group,electropuncture group and acupuncture group,the difference was statistically significant at T2(P<0.05);At T3,Comparison of T2,PWT and electropuncture group was significantly higher(P<0.05)than the acupuncture group.PWT model group was no significant difference at compared with T2(P>0.05).(5)Western blot results suggest that the control group p-JAK2 and p-STAT3 was the lowest,followed by electropuncture group group,cupuncture group was more than the amount of the aforementioned groups,but the highest expression in the model group.Conclusion:Eectropuncture may be through inhibition of the spinal cord JAK2 / STAT3 signaling pathway activation and inhibition of type 2 diabetes alleviate neuropathic pain. |
