Effect Of Blueberry On Expression Of Nqo1、GCLC、UCP2 In Non-Alcoholic Fatty Liver Disease In Rats

Objective:To investigate the effect of blueberry of NAD(P)H:quinine oxidoreductase 1(Nqo1)、Gamma glutamylcysteine synthetase catalytic subunit(GCLC)、Uncoupling Protein 2(UCP2) in rats and preliminary clear the mechanism of blueberry in oxidative stress injury in nonalcoholic fatty liver disease. Methods:Fifty healthy SD rats were randomly divided into five groups:normal control group,blueberry group,magnesium isoglycyrrhizinate group,model group and Geniposide group.The rats in normal control group were given normal diet, while other groups were all given high fat diet. At the same time, blueberry group were intragastrically given blueberry juice by gavage,when magnesium isoglycyrrhizinate group were given magnesium isoglycyrrhizinate solution,Geniposide group were given Geniposide solution and those in other groups were given physiological saline daily.At the end of week 12,all rats were sacrificed. Pathological changes in the hepatic tissue by HE staining or Oil-Red staining, the level of alanine aminotransferase(ALT), aspartate aminotransferase(AST), triglyceride(TG), total cholesterol(TC)and free fatty acid(FFA)in serum were detected by automatic biochemical analyze. The hepatic Glutathione Tablets(GSH) contents, Malondialdehyde(MDA)contents and Superoxide Dismutase(SOD)activity were quantified by ELISA. The ATP content of hepatic cell was detected by ADP/ATP luciferase method. The expression of Nqol and GCLC in liver tissue was detected by Western blot.The expression of UCP2 in liver tissue was detected by Western blot and Real-time PCR Results:Compared to model group,blueberry group liver and magnesium isoglycyrrhizinate group liver showed less pathological damage,serum ALT,AST,TG,TC and FFA levels decreased(P<0.05).The hepatic MDA also decreased(P<0.05),while GSH content and SOD activity increased(P<0.05). Compared to normal group, the expression of GCLC levels of model group increased(P>0.05), while the expression levels of blueberry group liver and magnesium isoglycyrrhizinate group increased(P<0.05).Compared to normal group, the expression of Nqol levels of model group increased(P<0.05), while the expression levels of blueberry group liver and magnesium isoglycyrrhizinate group also increased(P<0.05).The expression of UCP2 levels of blueberry group decreased(P<0.05),while the level of ATP increased(P<0.05). Conclusion:Blueberry possessed a protective effect on nonalcoholic fatty liver disease rats.The mechanism might be associated with up-regulating the expression of GCLC and Nqo1 protein of NAFLD liver;on the other hand,Blueberries could down-regulating the expression of UCP2 of NAFLD liver,increase the antioxidant capacity and energy synthesis of hepatic cell and inhibit NAFLD.
