| Purpose:This experiment mainly observe the effect of Jiedu Tongluo Baoshen Method to ILK and Akt of the foot cell of experimental DM rats' kidney,the effect of apoptosis and from lead to DN foot cell ILK-PI3 K PKB/Akt signaling pathway method to study the mechanism.Method:60 Wistar rats were randomly divided into: normal control group(group A)and model replication group.High sugar,high fat diet + one-time high-dose intraperitoneal injection of STZ on an empty stomach(50 mg/kg)building;divide the successful models into DM model group(group B),Irbesartan group(group C),Chinese medicine group(group D).Chinese traditional medicine group are given Jiedu Tongluo Baoshen powder to gavage administration1.25g/ml,4ml each day;Irbesartan group are given Irbesartan 17.5mg/kg to gavage;the normal control group and the DM model group are given distilled water as such as the volume of the other two groups.observe the general condition of rats,and at the 4th weekend and 8th weekend,test 24 h urine test,urine trace albumin.At the 8th weekend test FPG,Scr,BUN,weigh the left kidney weight,prepare kidney homogenate.ELISA method to detect ILK,AKT expression level in serumResults: Group C,D group compared with group B,Scr,BUN,kidney weight/body weight,urine,urine trace albumin were significantly lower,compared with group B,group D rats significantly lower fasting plasma glucose.Immune histochemical method detection results show that compared with group A,B,C,D group of experimental rats ILK expression increased,but the group C,group D expression in the B group were significantly lower;Compared with the rats in group A,group B in the rat kidney tissue in the expression of p-Akt significantly reduced;Compared with group B rats,group C,group D p-Akt expression in rat kidney tissue increased obviously.Conclusion:1.Jiedu Tongluo Baoshen Powder can reduce the DM rat kidney weight/body weight ratio,lower fasting plasma glucose,and improve the function of kidney of rats,reduce the urine trace albumin,have protective effect to the kidney.2.Preliminary clarify Jiedu Tongluo Baoshen Powder in DM rats foot protection mechanism of cell damage is effectively blocking ILK mediated PI3 K PKB/Akt signal transduction pathways,inhibiting cell apoptosis.3.This experiment to study the DN signal transduction pathological mechanism provides new ideas,new methods,and to provide prevention and control of traditional Chinese medicine from the perspective of regulating signal transduction DN new drug targets,and treatment. |
