Objective: To investigate the relationship between the autophagy gene Atg5 signaling pathway m TOR and COX-2 inhibitor nimesulide of NAFLD,reveal the nimesulide of steatosis liver cell mechanism of Autophagy.Methods:1、CμLtivate L-O2 cells in vitro, Using MTT method in combination with oil red o stain TG test to determine the best time and concentration of OA; Using MTT method to determine the best concentration of nimesulide and rapamy.2 、 Experiment was designed into 5 groups: normal control group, steatosis model group, nimesulide group, rapamy group and nimesulide rapamy group;Using Q-PCR method for detecting gene expression of m TOR and Atg5.Results:1、According to results of oil red o stain MTT and TG test determine the best OA NIM and RAPA concentration was 20μg/m L 200μg/m L and 30nmol/L,72 h was the best time of OA.2. There was lower m TOR gene expression of nimesulide group than steatosis model group(P<0.05).3. There was lower m TOR gene expression of nimesulide rapamy group than nimesulide group(P<0.05).4. There was higher autophagy gene Atg5 expression of nimesμLide group than steatosis model group(P<0.05).5. There was higher autophagy gene Atg5 expression of rapamy group than steatosis model group(P<0.05).Conclusions:1. The expression of NAFLD autophagy gene Atg5 was upregulated by cox-2 inhibitor nimesulide.2. The expression of NAFLD autophagy gene m TOR was downregulated by cox-2 inhibitor nimesulide.3. The expression of NAFLD autophagy gene Atg5 was upregμLated by cox-2 inhibitor nimesulide maybe through down-regulated m TOR signaling pathway gene of m TOR.
