| Objective:The purpose of this study was to investigate the expression of miR-200 c in serum and tissue of gastic cancer and analyze the correlation of miR-200 c expression in serum and tissues,so as to explore whether miR-200 c can be a serum marker for gastric cancer.Methods:(1)Tissue and peripheral blood samples were obtained from 30 cases of gastric cancer patients without radiotherapy and chemotherapy; and another 30 cases samples were obtained from healthy volunteers.( 2) The qRT-PCR was used to assess the relative expression of miR-200 c in each sample.( 3) Analyzing the expressional difference of miR-200 c between patient with gastric cancer and healthy control volunteer, the relationship between miR-200 c expression and clinical pathological parameters of gastric cancer, and the correlation of miR-200 c expression in serum and tissue.Results:(1)The expression of miR-200 c were 0.59±0.96,0.57±0.96 and 1.23±1.00 in serum, tumor tissue and adjacent tissue of gastric cancer on average; and they were 1.27±1.00 and 1.32±1.02 in serum and gastric tissue in healthy control volunteers. There was a statistical difference(p<0.05) about expression of miR-200 c in serum between gastric cancer and healthy control volunteer; and alsoa statistical difference( p<0.01) between the tumor tissue and adjacent gastric tissue; and steel a statistical difference(p<0.05) in tumor tissue and normal gastric tissue; while no statistical difference(p>0.05) between adjacent gastric tissue and normal gastric tissue.(2)The expression of miR-200 c taken on a significant positive correlation between serum and tissue of gastric cancer(r=0.996,p<0.01);and also a significant positive correlation between serum and tissue of healthy control volunteers(r=0.994,p<0.01).(3)The expression of miR-200 c were 0.95±0.68 and 0.25±0.32 in stage ofⅠ~Ⅱand Ⅲ~Ⅳon average, and there was a statistical difference(p<0.05) between them.While it was not significantly correlated with the patients’ age, differentiation degree,size of tumor and lymph node metastases(p>0.05).Conclusions:(1) miR-200 c is low expression in serum and tissue of gastric cancer, acting as a tumor suppressor gene.(2) The expression of miR-200 c taken on a significant positive correlation between serum and tissue of gastric cancer,thus, in theory, miR-200 c can be used as a serum marker for gastric cancer.(3)The expression of miR-200 c in serum were relative to the stage of TNM, the later stage, and the lower expression of miR-200 c. While it was not significantly correlated with the patients’ age, differentiation degree,size of tumor and lymph node metastases. |
