Objective:To study basil polysaccharide extraction, purification and physicochemical properties of the basic. Discuss basil polysaccharide combined with paclitaxel inhibit the tumor-bearing H22 Mice.To observe whether it can improve ascites tumor H22 Mice the survival rate and improve the quality of life.Methods: Extract basil polysaccharide by water extraction and alcohol precipitation method,Measuring Basil crude polysaccharide content by phenol sulfuric acid method,Measuring protein content by Kaumas blue.Removing protein by sevag method,Removing small molecular impurities such as pigment by dialysis,The polysaccharide was isolated and purified by DEAE-52 ion exchange column chromatography.Pre column derivatization GC-MS spectrometry for the determination of polysaccharide of monosaccharide composition.By detecting H22 tumor bearing mice tumor weight and survival rate, Observe basil polysaccharide combined with Taxol synergistic effect on H22 tumor-bearing mice.Through the detection of serum ALT, AST and urea nitrogen,To observe the effect of basil polysaccharide combined with medicine on H22 ascites tumor mice liver and kidney.Results: basil polysaccharide is 28% measured by phenol sulfuric acid method,The protein content is10% measured by Kaumas blue,Removing protein by sevag method,Purification rate was 60%.Purification by DEAE chromatography column,Neutral polysaccharide and acid polysaccharide were obtained,The results were homogeneous polysaccharide measured by agarose gel electrophoresis.basil polysaccharide is composed of glucose, galactose, mannose, rhamnose, fucose and arabinose. Molar ratio is:2.33:2.4:1:1.34:2.22:1.96.basil polysaccharide can increase the antitumor effect of paclitaxel. Paclitaxel 10mg/kg+ Basil polysaccharide 100 mg / kg group tumor inhibition rate is higher 30.94% compared with paclitaxel 10mg/kg group. basil polysaccharide combined with paclitaxel can improve the survival rate of mice and have no damage to liver and kidney.Paclitaxel 10mg/kg+Basil polysaccharide 100mg/kg group survival rate is higher 58.2% compared with paclitaxel 10mg/kg group.Conclution: Water extraction and alcohol extraction yield of basil polysaccharide extraction precipitation method has higher extraction rate.saving energy. It can be used as the common extraction method of basil polysaccharide.Sevage method for protein removal,The loss of polysaccharide was small, and the purification rate was high.The pharmacodynamics experiment proved that basil polysaccharide combined with paclitaxel can inhibit the growth of tumors,and improve the survival rate of mice, It prove that basil polysaccharide has greater research space in antit-umor applications.
