Objective: To observe anti-inflammatory and analgesic effects of modified Xiaochaihu decoction by establishing the model of Inflammation and pain in mice,and comparing with Yinqiao powder and paracetamol,to provide experimental basis for the modified Xiaochaihu decoction clinical application.Methods: 84 healthy kun ming mice were divided into 7 groups by using weight stratified random digital look-up table method,and then marked.Each group is 12.the mice were divided into blank group,model group,Yinqiao powder,paracetamol,modified Xiaochaihu decoction high,medium and low dose group.Adaptive raising them three days,and weighing,continuous lavaging for 3 days,lavaging the blank group,model group lavage with saline water,twice one day,20 ml/kg capacity.The dose lavage was 28.08 g/kg every day to the Yinqiao powder group,the acetaminophen group was 0.78 g/kg,the modified Xiaochaihu decoction high,medium and low dose group were 33.15g/kg,baijie19.89g/kg,6.63g/kg.After lavaging the mice 30 minutes the seventh day,smearing the formaldehyde to the toes of the mice,observing the add foot behavior of the mice,and recording add foot number every group of mice within 10 minutes.lavaging the mice 30 minutes the eighth day,injecting the enterocoelia of mice with 0.6% acetic acid by dose of 0.2 ml/one to cause pain,recording animal number of mice writhing reaction and the number of twist body within 10 min after injection of acetic acid.After lavaging the mice 30 minutes the ninth day,smearing the positive and negative right ear of the mice evenly by 40?l xylene to cause inflammation.The left ear was comparison.Sampling blood by removing eyebolls after 30 minutes,Collecting serum after centrifugation,Testing groups of mice serum PGE2,IL-1? content.The results were statistically processed column chart.After picking blood,killing off cervical vertebra in mice,Cutting out two ear in mice,laying a circular meat respectively with a diameter of 9 millimeter ear swelling of ear device in the same place,weighing by a analytical balance.Putting the left and right ear piece weight difference as inflammatory swelling degree index,Calculating the ear swelling degree and the inhibition rate.Using SSPS17.0 statistical analysis software for data processing.Result:1)The mean of adding foot behavior of every group mice was lower than the model group except the paracetamol group.The mean of twisting body behavior of every group mice was lower except the modified Xiaochaihu decoction high concentration group(P<0.05).The mean of adding foot behavior of every group mice was lower than the model group except the paracetamol group(P<0.01).The number of adding foot behavior of Yinqiao powder group and modified Xiaochaihu decoction(low,mid,high concentration groups)were lower than paracetamol group(P<0.01).The number of modified Xiaochaihu decoction high concentration group was lower than Yinqiao powder group(P<0.01).The number of writhing times of modified Xiaochaihu decoction high concentration group was lower than Yinqiao powder group and paracetamol group(P<0.01).2)Compared with the model,the experimental mice serum IL-1?,PGE2 levels were lower(P< 0.01);The IL-1?in serum of modified Xiaochaihu decoction(medium and high concentration groups)was lower than the paracetamol group(P < 0.01).The IL-1?in serum of modified Xiaochaihu decoction(low,medium and high concentration groups)was lower than the Yinqiao powder group(P< 0.01);The PGE2 in serum of modified Xiaochaihu decoction(medium and high concentration groups)was lower than the paracetamol group(P< 0.01);The PGE2 in serum of modified Xiaochaihu decoction low concentration group was higher than the acetaminophen group(P< 0.05),but lower than Yinqiao powder group(P< 0.01).The PGE2 in serum of modified Xiaochaihu decoction high concentration group was lower than Yinqiao powder group(P< 0.01).3)The mice auricle swelling around variation was larger,no statistical significance.4)The weight of the mouse were not big change because of the short time.Groups of experimental index has little relationship between gender differences.Conclusion:Paracetamol,Yinqiao powder had a positive impact to the mice on pain stimulation,immunology index.The anti-inflammatory effect of high,medium concentration modified Xiaochaihu decoction were stronger than paracetamol and Yinqiao powder.The reduction analgesic effect of modified Xiaochaihu decoction group(high and mid concentration groups)were stronger than paracetamol,The reduction analgesic effect of modified Xiaochaihu decoction group high concentration group was higher than that of Yinqiao powder,The later was stronger than the low concentration modified Xiaochaihu decoction.It Indicates that modified Xiaochaihu decoction had certain advantages of anti-inflammation and analgesia.It provided the experimental basis to the modified Xiaochaihu decoction on the anti-inflammation and analgesic action.
