Objective:Observing the Influence of Houpu Mahuang Decoction on lung tissue pathology and the contents of IL-13、INF-γ of asthmatic,approaching the mechanism of action of Houpu Mahuang Decoction on treating bronchial asthma,it can provide laboratory evidence for the future clinical application.Methods:Choosing 60 Female Kunming mice which were weighed and labeled and randomly divided into 6 groups and 10 mice in each group.Specific groups as follows: normal group(group A);asthma model group(group B);control group(Group C);the Houpu Mahuang Decoction low dose group(Group D);the Houpu Mahuang Decoction dose group(Group E);Houpu Mahuang Decoction high dose group(Group F).Using the egg protein and aluminum hydroxide stimulate sensitization and asthma mice model with the method of replication ova induced excitation stage,that could be successfully produced in mice with asthma.mice in each group were given corresponding medicine intervention,the experimental process and detailed record of mice to observe changes in overall situation.after completion of the experiment,observed under light microscope the pathological changes of lung tissues of mice,and detected the changes of IL-13 and INF-γ in the serum of mice in ELISA method.The related experimental data were analysed using SPSS18.0.Results:1.The treatment group could significantly improve the general state of mice.Around the asthma model group mice bronchial wall and the tube wall had a large number of eosinophils and lymphocytes infiltration of inflammatory cells,bronchial mucosa edema,thickening,cilia lodging,deletion,epithelial shedding,mucus secretion increased.Compared with asthma group the pathological changes of Houpu Mahuang Decoction dose group and control group were not obvious,Houpu Mahuang Decoction high dose group and dexamethasone group was significantly improved,Houpu Mahuang Decoction low dose and middle dose group improved than dexamethasone group.2.Compared with the control group,the asthma model group,Houpu Mahuang Decoction dose group and the control group,the content of IL-13 increased,the difference was significant(P < 0.01);Houpu Mahuang Decoction dose group and the control group,the content of IL-13 lower than that of asthma model group(P < 0.01);Houpu Mahuang Decoction in the low,middle dose group,the content of IL-13 higher than high dose group and the control group(P < 0.05).Houpu Mahuang Decoction high dose group and the control group close,with no significant difference(P > 0.05);Houpu Mahuang Decoction low group and the middle group close,with no significant difference(P > 0.05).3.Compared with the blank control group,asthma model group,Houpu Mahuang Decoction dose group and the control group of INF-γ level decreased significantly(P < 0.01);Houpu Mahuang Decoction dose group,control group and the asthma model group compared with significant difference(P < 0.01);Houpu Mahuang Decoction low dose group and middle dose group and dexamethasone group(P < 0.05);Houpu Mahuang Decoction low dose group and middle dose group and high dose group(P < 0.01).Houpu Mahuang Decoction high dose group and dexamethasone group(P > 0.05)compared.Conclusion: Asthma model of mice lung tissue pathologic change,also model mice appear a series of typical asthma performance,verify the modeling method is feasible;the Houpu Mahuang Decoction can improve the overall symptoms of asthma mice,and obviously improve the reaction degree of airway inflammation in a mouse model of asthma;The pathological changes of Houpu Mahuang Decoction can improve the lung tissue of asthmatic mice.Houpu Mahuang Decoction can significantly decrease the IL-13 level in serum of asthmatic mice,elevated INF-γ levels.Houpu Mahuang Decoction can reduce the level of IL-13 and increasing the level of INF-γ,Th1 and Th2 ratio imbalance adjustment,so as to achieve the purpose of treating asthma.
