Objective:To analysis the effects of insunlin secretion by1-methylhydantoin with INS-1 cell and its possible mechanismMethods:INS-1 cell in vitro cultured rat insulin tumor cell lines.To deal them with 1-methylhydantoinand octreotide with different concentration in 72 hours.Detected INS-1 cell’s proliferation rate with CCK-8 method.Radioimmunoassay used to detect the content of insulin of each group.FCM detect apoptosis and cell cycle;Western Blot detect the possible mechanism.Results:1.CCK-8 results: 1-methylhydantoin and octreotide can suppress INS-1 cell’s proliferation rates,,and their intensity of inhibition rate depended on concentration.2. Radioimmunoassay results: 1-methylhydantoin and octreotide can suppress insulin secretory,and their intensity of inhibition rate depended on concentration.3.FCM assay results:Compared with control group, there were significant differences of apoptosis rate and cell cycle of INS-1 cells in each treatment group of 1-methylhydantoin and octreotide.4.Western Blot results:Compared with the control, 1-methylhydantoin and octreotide treated INS-1 cell,the cAMP and PKA protein obvious reduce.Conclution:1. 1-methylhydantoin can inhibit the proliferation of INS-1cell. Its possibly mechanism might be related on the cell apoptosis and the cell cycle block.2. 1-methylhydantoin can inhibit the secretion of insulin in INS-1 cell. Its possibly mechanism might be related to the inhibition of the expression of cAMPPKA pathway.
