The objective of this study is testing the expression of miR31、miR 125b、miR 328 and the target genes of RASA1、MMP13、PAK6, and exploring the carcinogenic mechanism of miRNA in CSCC, then the result will establish the stable theory basea of the targeted therapy for CSCC.Methods:Specimens which were 30 cases of fresh biopsies of cutaneous squamous cell carcinoma and 30 cases of normal skin tissue come from the department of dermatology and department of plastic surgery in Zhongda Hospital affiliated to Southeast University during 2014-2016, saved in the refrigerator of -80 ℃. All specimens were done HE stain; we use RT-PCR to test the expression of miR31、 miR125b、miR328、RASA1、MMP13 and PAK6 in CSCC specimens; Using the Pearson correlation analysis to test the correlation of the miR31 and RASA1、 miR125b and MMP13、miR328 and PAK6.Results:1 The expression of miR31、miR125b and miR328 in CSCC specimensThe average relative expression of miR31 in cases and normal group respectively were 0.951±0.605 and 0.514±0.521. The average relative expression of miR31 in CSCC was higher, There was a significant difference between the two groups (P<0.001).The average relative expression of miR125b in cases and normal group respectively were 0.460±0.362 and 0.428±0.327.There was no significant difference between the two groups (P>0.05).The average relative expression of miR328 in cases and normal group respectively were 0.066±0.087 and 0.125±0.112. The average relative expression of miR328 in CSCC was lower, There was a significant difference between the two groups (P<0.05).2 The expression of RASA1、MMP13 and PAK6 in CSCC specimens and normal skin tissueThe average relative expression of RASA1 in cases and normal group respectively were 0.775±0.871 and 1.540±1.787. The average relative expression of RASA1 in CSCC was lower, There was a significant difference between the two groups (P <0.05).The average relative expression of MMP13 in cases and normal group respectively were 0.013±0.069 and 0.002±0.002.There was no significant difference between the two groups (P>0.05).The average relative expression of PAK6 in cases and normal group respectively were 0.122±0.127 and 0.041±0.082. The average relative expression of PAK6 in CSCC was higher, There was a significant difference between the two groups (P<0.05).3 The correlation between miR-31 expression and RASA1 expression in CSCC, the correlation between miR-328 expression and PAK6 expression in CSCC.In CSCC, the result of the correlation between miR-31 expression and RASA1 expression show that r=-0.407,P=0.026, prompting there is a negative correlation between miR-31 expression and RASA1 protein expression in CSCC. the result of the correlation between miR-328 expression and PAK6 expression show that r=-0.389,P =0.034, prompting there is a negative correlation between miR-328 expression and PAK6 protein expression in CSCC.Conclusion:1. The expression level of miR31 is higher in CSCC. We consider miR31 is CSCC-associated MicroRNAs, the expression level of target gene of RASA1 is lower in CSCC, there is a negative correlation between miR-31 expression and RASA1, the high expression of miR31 may promote the proliferation of cells through down-regulation of RASA1 protein expression;2. The expression level of miR328 is lower in CSCC. We consider miR328 is CSCC-associated MicroRNAs, the expression level of target gene of PAK6 is higher in CSCC, there is a negative correlation between miR-328 expression and PAK6, the lower expression of miR328 may promote the proliferation of cells through up-regulation of PAK6 protein expression;3. The expression level of miR125b and MMP13 has no significant difference between the test and control cases.
