Objective To investigate the expression of Stat3、GABA(A)Rα1, and the relative m RNA level of ICER(Stat3-regulate gene)、GABRA1(GABA(A)Rα1 encoding gene) in low grade glioma(LGG) and peritumoral tissue. And to explore their relationship with LGG-associated epilepsy.Methods To collect postoperative brain tissue specimens of 42 patients, LGG(n=31) and peritumoral tissue(n=20) with epilepsy, LGG(n=11) and peritumoral tissue(n=9) without epilepsy, normal brain tissue(n=8).To compare their expression of Stat3、GABA(A)Rα1,and relative m RNA level of ICER、GABRA1 by Immunohistochemistry and RT-PCR.Results 1.Immunohistochemical staining showed that Stat3 was mainly distributed in the nucleus and cytoplasm, GABA(A)Rα1 was in the cell membrane and cytoplasm. Compared with control group, the expression of Stat3 in glioma tumor and peritumoral tissue was higher, while the expression of GABA(A)Rα1 was lower(P < 0.05). We also found that the expression of Stat3 in peritumoral tissue with epilepsy was higher than without epilepsy, and the expression of GABA(A)Rα1 in peritumoral tissue with epilepsy was lower than without epilepsy(P < 0.05). 2.RT-PCR showed that the relative m RNA level of ICER in LGG and peritumoral tissue was higher than the control group, while the relative m RNA level of GABRA1 is lower(P < 0.05). We also found that the relative m RNA level of ICER in peritumoral tissue with epilepsy was higher than without epilepsy, and the relative m RNA level of GABRA1 in peritumoral tissue with epilepsy was lower than without epilepsy(P < 0.05).Conclusions 1.The higher expression of Stat3 and lower expression of GABA(A)R α1 in peritumoral tissue is relative with LGG-associated epilepsy. 2.Epileptogenic zones in LGG exists Stat3 signaling pathways activation, and Stat3 may through regulating GABA(A)Rα1 participated in the low grade gliomas correlation development of epilepsy.
