Objective: To explore how clopidogrel attenuates acute lung ischemia-reperfusion in mice model.Methods: Animals were divided into sham operation or ischemia reperfusion. Clopidogrel was delivered once a day(20mg/kg) for 3 days before surgery in Sham+ Clo and IR+Clo groups.While the sham and IR groups received an oral gavage administration with sterile water(the same volume as Clo). The left lungs were harvested 120 min post procedures.The histological changes of harvest left lung were analyzed.The lung wet-to-dry weight ratio,Myeloper-osidase(MPO) activity,IL-6, TNF-? were measured.The flow cytometric analyses were employed to detect the activation of platelets and leukocytes,platelet-leukocyte coaggregates.Results: There are no significant differences between Sham groups and Sham+Clo groups. Compared with Sham groups, lung W/D ratio,MPO activity and the level of IL-6 and TNF-? in IR groups were significantly enhanced. The expressiong of CD61 on platelets and CD11 b on leukocytes,the percentages of platelet-leukocyte coaggregates in IR groups were also enhanced.Clo significantly attenuates LIRI,reducing lung W/D ratio,MPO activity and the level of IL-6 and TNF-?.Compared with IR groups,the expressiong of CD61 on platelets and CD11 b on leukocytes,the percentages of platelet-leukocyte coaggregates in Clo group were significantly reduced.Conclusion: Clopidogrel significantly attenuates acute lung ischemia-reperfusion injury in mices.The mechanism may involve in inhibiting the activation of platelets and leukocytes,the interaction of plateles with leucocytes and the expression of TNF-?,IL-6.
