Objectives To analyze the expression of Omi/Htr A2(mitochondrial serine proteas e) in papillary thyroid carcinoma,nodular goiter and normal tissues,and its correl ation with the clinicopathologic parameters of papillary thyroid carcinoma.Methods 1 Collecting 120 cases of papillary thyroid carcinoma, 40 cases of nodular goiter and 40 cases of normal tissues in the Workers hospital of Tangshan from February2013 to October 2014. In 120 cases of papillary thyroid carcinoma, 52 cases of male and68 cases of female; 56 cases of less than 45 years old,64 cases of more than 45 years old;35 cases of with extracapsular invasion, 85 cases of without extracapsular invasion; 42 cases of patients with low degree of differentiation, 78 cases of patients with high and middle differentiation degree;According to the TNM(T shows the size of the primary tumor, N shows the regional lymph node metastatic, M shows distant metastasis) of UICC(International Union of counteracting cancer)in 2002, We stage these papillary thyroid carcinoma tissues, 65 cases of Ⅰ~Ⅱperiod, 55 cases ofⅢ~Ⅳperiod; 70 cases of with lymph node metastatic, 50 cases of without lymph node metastatic.2Immunohistochemistry and Western blotting were used to assess the expression of Omi/Htr A2 protein in 120 cases of papillary thyroid carcinoma, 40 cases of nodular goiter and 40 cases of normal tissues,and its association with clinicopathologic parameters were analyzed.Results 1 The positive expression of Omi/Htr A2 in the thyroid tissues appear pale yellow, brown yellow or brown. The positive expression rates of Omi/Htr A2 in papillary thyroid carcinoma, nodular goiter and normal thyroid tissue were 71.67%(86/120), 30%(12/40) and 32.50%(13/40). The expression of Omi/Htr A2 was positive in papillary thyroid carcinoma tissue, which was significantly higher than that in nodular goiter and nomal tissue(χ2 = 21.944,19.509,P < 0.05). The expression of Omi/Htr A2 was no significant difference in the group of the nodular goiter and the normal thyroid(χ2=0.058,P > 0.05).2 Western blotting: the relative amount of Omi/Htr A2 protein in papillary thyroid carcinoma tissue was1.184±0.060, significantly higher than in nodular goiter(0.599 ± 0.096) and normal tissues(0.606 ± 0.097)(P < 0.05). The relative amount of Omi/Htr A2 protein was no significant difference in the group of the nodular goiter and the normal thyroid(P>0.05).3 The expression of Omi/Htr A2 wascorrelated with extracapsular invasion,lymph node metastasis, clinical staging and differentiation(P < 0.05). The positive expression rates of Omi/Htr A2 in papillary thyroid carcinoma without extracapsular invasion was hihher than that of papillary thyroid carcinoma with extracapsular invasion(χ2=34.003,P< 0.01); The positive expression rates of Omi/Htr A2 in papillary thyroid carcinoma without lymph node metastasis was hihher than that of papillary thyroid carcinoma with lymph node metastasis( χ2=6.421, P< 0.05); The positive expression rates of Omi/Htr A2 in papillary thyroid carcinoma inⅠ ~ Ⅱperiod was hihher than that of papillary thyroid carcinoma in Ⅲ~Ⅳperiod(χ2=11.710,P< 0.01); The positive expression rates of Omi/Htr A2 in papillary thyroid carcinoma with high differentiation was hihher than that of papillary thyroid carcinoma with low differentiation(χ2=30.957,P< 0.01); The expression of Omi/Htr A2 was not correlated with gender,age, whether complicated with Hashimoto’s thyroiditis and location(P>0.05).Conclusions 1 Omi/Htr A2 protein was overexpressed in papillary thyroid carcinoma.2The expression of Omi/Htr A2 was correlated with extracapsular invasion,lymph node metastasis, clinical staging and differentiation.3 The abnormal expression of Omi/Htr A2 can be used as a new marker for the diagnosis of thyroid papillary carcinoma, and may be a new target for the treatment of thyroid papillary carcinoma.
