| Objective: The incidence of intrahepatic cholangiocarcinoma(ICC) is on the increase each year. ICC has been characterized by difficult diagnosis at the beginning, highly metastatic capability, insensitivity to chemotherapies and poor prognosis. As such, less is known about the diagnosis marker,therapy and prognosis standard of ICC at this moment. Here we analysed the expression difference of long non-coding RNA(Lnc RNA) in ICC tumor tissue and the function of Lnc RNA in ICC pathogenesis to identify the Lnc RNAs related to ICC incidence, treatment and prognosis.Methods: Both normal and ICC tumor tissue(confirmed by clinical pathology) samples were collected for analysis. To identify the Lnc RNAs related to tumor, Angilent 4*180k Human Lnc RNA Chip was used to screen the expresson of Lnc RNA and m RNA,and co-expression analysis of m RNA-Lnc RNA was also used to predict potential function of Lnc RNA.Results: The results of microarray screening showed that, compared with normal tissue,718 m RNAs exhibited expression difference in tumor including 432 upregulation and 286 downregulation. Additionally, there were 1740 Lnc RNAs whose expression level changed and 1427 of them were increase in tumor, but other 313 Lnc RNAs were decreased. Among them, 86 Lnc RNAs were found to express differently(p<0.01) and the fold of expression difference was more than 4,and Lnc RNA CRNDE/ CBR3-AS1/SOX2-OT that relevant to tumor was differential expression in ICC.Conclusion: Our results may suggest that Lnc RNA is involved in cellular function in various aspects leading to their important roles in pathogenesis of ICC by regulate the intracellular signaling pathway. Lnc RNA CRNDE/CBR3-AS1 may have a key role in the pathogenic process of ICC.Also,signaling pathway of Mapk/Rap1/Ras/PI3K-Akt may play an important role in the pathogenesis of ICC. |
