Objective: To investigate the effects and mechanism of action of Regulating Collateral and Eliminating Turbidity Granules(REG) on micro RNA-200 a and mi R-29 c expression in renal interstitial fibrosis in a rat, and explore the application of mediation method in renal interstitial fibrosis.Methods: SD mice were randomly divided into three groups(10 mice/group), including sham group, UUO group, and UUO+ REG group The model was established by UUO, as previously reported. Briefly, UUO group and UUO+ REG group animals were placed under general anesthesia by a peritoneal injection of 10% chloral hydrate(0.35mg/kg) solution. Complete ureteral obstruction was induced by ligation of the left ureter with 5-0 silk at the renal calyx and lower renal pole. Sham animals were also placed under general anesthesia and the abdomen was cut open to liberate the ureter without subsequent ligation. On the first day after operation, UUO+REG animals were perfused with REG by intragastric administration at the dose(10g/kg.d-1), while Sham and UUO animals were perfused with normal saline. Perfusions were repeated on days 7 and 14, post-operation. Animals were euthanized either on days 7 or 14 post-operation, and the left kidney was removed and cut along their coronal section. subsequently examined for changes in morphology or for changes in α-SMA, kidney were observed by hematoxylin-eosin staining, and quantitative RT-PCR for micro RNA-200 a and micro RNA-29 c expression.Results: The expression of α-SMA increased significantly(P < 0.01) in both UUO and UUO+REG groups at 7 d and 14 d post-operation; however, there was a significant difference in α-SMA expression in UUO+REG group compared with UUO group(P < 0.05). the expression levels of mi R-200 a and mi R-29 c were reduced in both UUO and UUO+REG group as compared with that in Sham group at 7 d and 14 d post-operation. At 7 d and 14 d post-operation, the expression of mi R-200 a in UUO group was significantly lower than that in Sham(P<0.01), andthat expression in UUO+REG group was different from the others(<0.05).The level of mi R-29 c was reduced in both UUO and UUO+REG groups as compared with that in Sham group at 7 d and 14 d post-operation(P<0.05), while the expression in UUO group and UUO+REG group were lower than that in Sham(P<0.05); and there was no difference between UUO group and UUO+REG group; at 7d post-operation, but UUO group was lower than the other two groups(<0.05).Conclusion: REG could delay the progression of chronic renal failure by up-regulating mi R-200 a and mi R-29 c.
